两种α-糖苷酶抑制剂体外筛选模型的改良与比较

Modification and Comparison of Two in vitro Assay Methods for Screening α-Glucosidase Inhibitors Produced by Lactic Acid Bacteria

摘要: α-糖苷酶抑制剂可以控制餐后血糖浓度波动,从而达到预防和治疗糖尿病的作用。本文对两种α-糖苷酶抑制剂体外筛选模型进行了改进。结果表明,以阿卡波糖作阳性对照,两种模型均适用于以脱脂乳为生长介质的产α-糖苷酶抑制剂的乳酸菌的筛选。选取12株特定乳酸菌分别用两种模型测其发酵脱脂乳上清对α-糖苷酶的抑制活性,两种模型筛选结果一致,但测得的抑制活性呈现一定的差异。当加入到脱脂乳中的阿卡波糖浓度较低时,以麦芽糖为底物的酶-抑制剂模型测得的α-糖苷酶抑制活性高于以4-硝基酚-α-D-吡喃葡萄糖苷(PNPG)为底物的酶-抑制剂模型。

Abstract: α-Glucosidase inhibitors are therapeutic agents for reducing postprandial hyperglycemia,which are widely applied in clinics for prevention and management of type II diabetes mellitus.In this study,two in vitro assay methods were modified to adopt the screening α-glycosidase inhibitors (α-GI) produced in reconstituted skimmed milk by lactic acid bacteria,enrolling acarbose as a positive control.Both of the modified in vitro assay methods were proved to be suitable for screening α-GI produced by lactic acid bacteria,with the half inhibitory concentration (IC50) for acarbose of 0.6056 mg/mL obtained by PNPG method and 0.3843 mg/mL by maltose α-glycosidase method respectively.The inhibitory effects of 12 selected lactic acid bacteria strains were consistent between the PNPG and the maltose α-glycosidase method,although the inhibitory rates measured by the latter method were slightly higher.When in low concentration,the inhibitory rates of acarbose in reconstituted skimmed milk obtained by maltose α-glycosidase (invertase) method were higher than those by PNPG method.
