蚯蚓葡萄糖氧化酶的提取纯化研究

摘要: 本研究采用闪式提取技术提取蚯蚓葡萄糖氧化酶(GOD)。利用正交设计优化实验条件,得到最优条件为:固液比1:4.5(g/mL),闪提时间45 s,超声时间30 min。通过酸沉淀,羧甲基纤维素CM-22离子交换层析和DEAE离子交换层析实现了GOD的纯化,回收率达到了60.8%,比活达到了2684.2 mU/mg,纯化倍数达到37.2倍。

Abstract: In this study,flash extraction was used to extract glucose oxidase (GOD) from earthworm.The extraction conditions were optimized by orthogonal test design and the optimal conditions were applied as follows:solid to liquid ratio of 1:4.5 (g/mL),flash extraction for 45 s and ultrasonic extraction for 30 min.GOD was purified by column chromatography on CM-22 and DEAE.The purified GOD had a specific activity of 2684.2 mU/mg with a yield of 60.8%.
