HPLC同时测定延胡索中6个核苷成分含量

Simultaneous Determination of 6 Nucleoside Components in Corydalis yanhusuo W.T.Wang by High Performance Liquid Chromatography

摘要: 以高效液相色谱法测定延胡索中6个核苷成分的含量,建立了延胡索中6个核苷胞苷、尿苷、腺苷、2-脱氧腺苷、鸟苷、胸苷成分的控制指标。采用Grace Prevail C18(4.6 mm×250 mm,5 μm)柱,流动相为甲醇(A)0.1%磷酸水溶液(B),流速为0.8 mL/min,柱温为30 ℃,检测波长为260 nm。结果胞苷、尿苷、腺苷、2-脱氧腺苷、鸟苷、胸苷在质量浓度分别在6.70~134.06、9.08~181.68、9.68~193.52、2.11~42.12、6.80~135.94和2.56~51.26 μg/mL范围内与峰面积呈良好线性关系(r≥0.9994);平均回收率(n=6)为 99.7%~102.1%,RSD≤ 2.3%,南通产区6个核苷含量分别为:20.97、207.10、221.01、6.82、152.52、11.60 μg/g,杭州产区含量为:21.58、207.69、220.70、8.37、154.20、11.52 μg/g,宜兴产区为:20.49、208.62、220.93、8.59、157.27、11.38 μg/g。本试验所建立的用于同时测定延胡索中6个核苷含量的分析方法简单、重复性好、准确高,可为延胡索的质量控制提供依据。

Abstract: To establish a HPLC method for the simultaneous determination of 6 nucleoside components in Corydalis yanhusuo W.T.Wang.The assay was performed on a Grace Prevail C18 (4.6 mm×250 mm,5 μm) column;mobile phase consisted of methanol (A)-0.1% orthophosphoric acid water (B) with gradient elution at the flow rate of 0.8 mL/min;column temperature was 30 ℃;and the detection wavelength was set at 260 nm.The investigated compounds,including cytidine,uridine, adenosine, 2-deoxyadenosine,guanosine and thymidine,showed good linearity (r≥0.9994) in the range of 6.70-134.06, 9.08-181.68, 9.68-193.52, 2.11-42.12, 6.80-135.94 and 2.56-51.26 μg/mL,respectively.The average recoveries (n=6) were within 99.7%-102.1% with RSD≤2.3%.The contents of 6 nucleoside components in C.yanhusuo samples from Nantong city were:20.97,207.10,221.01,6.82,152.52 and 11.60 μg/g;in C.yanhusuo samples from Hangzhou city were:21.58,207.69,220.70,8.37,154.20 and 11.52 μg/g and in C.yanhusuo samples from Yixing city were: 20.49,208.62,220.93,8.59,157.27 and 11.38 μg/g.The developed HPLC method was simple and valid.It can be used for quality evaluation and quality control of C.yanhusuo.
