南方大斑蝥体内结合斑蝥素对人肝癌HepG2细胞凋亡的影响

Effects of Bound Cantharidin from Mylabris phalerata Pallas on the Apoptosis of Human Hepatoma HepG2 Cells

摘要: 利用水提法从来自罗甸县茂井镇的南方大斑蝥中提取得到结合斑蝥素,作用于人肝癌HepG2细胞后,分别利用Annexin V-FITC/PI双染法检测HepG2细胞的凋亡率,RT-PCR法检测Bax、Bcl-2 mRNA相对表达情况,利用Western blot法检测HepG2细胞中Bax、Bcl-2蛋白表达的影响。实验结果表明罗甸县茂井镇产的南方大斑蝥体内结合斑蝥素可诱导人肝癌HepG2细胞凋亡,其机制可能与上调Bax和下调Bcl-2表达有关。

Abstract: Bound cantharidin was obtained from Mylabris phalerata Pallas (Maojing town of Luodian County) by water extraction method.The HepG2 cells were cultured in vitro with different concentrations of bound cantharidin.Then the annexin V-FITC/PI staining method was applied to detect the effect of bound cantharidin on apoptosis of HepG2 cells,and the expression of Bax,Bcl-2 gene in HepG2 cells were analyzed by RT-PCR.The protein expression of Bax,Bcl-2 in HepG2 cells were also detected by Western blot assay.The experimental results showed that bound cantharidin from M.phalerata-induced apoptosis of HepG2 cells,the mechanism behind may be connected with both the up-regulation expression of Bax gene and the down-regulation expression of Bcl-2 gene.
