LC-MS/MS法测定脑透析液中3种苯乙醇苷类成分的浓度及其体内外探针回收率考察

Determination of Three Phenylethanoid Glycosides in Intracerebral Dialysate by LC-MS/MS and Comparison of in vivo and in vitro Recovery of Microdialysis Probe

摘要: 本文采用LC-MS/MS联用法校正3种苯乙醇苷类成分(连翘酯苷B、毛蕊花糖苷、异毛蕊花糖苷)的体内外回收率,为微透析技术用于苯乙醇苷类成分的相关取样以及该类成分的微量检测提供实验基础。实验结果表明,连翘酯苷B、毛蕊花糖苷、异毛蕊花糖苷的体外回收率高于体内回收率,体外平均回收率分别为19.5%、27.5%、31.9%,体内平均回收率分别为11.6%、16.0%、16.9%。本实验利用大鼠脑内微透析采样技术,建立了同时检测多个苯乙醇苷类成分的LC-MS/MS方法,具有活体在线、实时取样,分辨率和灵敏度高,分析时间短等优点,适用于高通量生物样品的测定分析。

Abstract: To establish a method for detecting micro-dialysis recovery of forsythoside B,acteoside and isoacteoside,measuring the concentration of three phenylethanoid glycosides in intracerebral dialysate by LC-MS/MS method,and compare the probe recovery in vivo and in vitro.The recovery of three phenylethanoid glycosides was 19.5%,27.5%,31.9% in vitro and11.6%,16.0%,16.9% in vivo.The probe recovery of three phenylethanoid glycosides in vivo is lower than in vitro,and both are stable in different concentrations.The LC-MS/MS method is accurate,sensitive,and reproducible for quantitative determination of three phenylethanoid glycosides in microdialysate.
