大孔吸附树脂分离纯化党参中苍术内酯I和苍术内酯II的工艺研究

Enrichment of Atractylenolide I and Atractylenolide II from  Codonopsispilosula with Macroporous Resin

摘要: 采用大孔吸附树脂分离纯化党参药材中的主要活性成分苍术内酯I和苍术内酯II。以吸附量、吸附率、解吸附量、解吸附率为指标,筛选出较好的DM130型大孔树脂,并对苍术内酯I和苍术内酯II分离纯化的工艺条件进行了研究。结果表明用DM130型大孔吸附树脂对党参中的苍术内酯I和苍术内酯II进行分离纯化的最佳工艺条件为:30 ℃,供试品上样液的浓度为0.125 g/mL,上样液体积为200 mL,上样液流速为6 mL/min,95%乙醇作洗脱剂,洗脱剂流速为4 mL/min。经过纯化后,苍术内酯I和苍术内酯II的含量分别增加了32.3倍和25.6倍,表明DM130型大孔吸附树脂对党参中的苍术内酯I和苍术内酯II进行分离纯化的方法可行、有效。

Abstract: The purification process of atractylenolide I and atractylenolideII from Codonopsispilosula were studied.Adsorption capacity,desorption capacity,adsorption rate and desorption rate were selected as evaluation indexes,the types of macroporous resin was screened and the purification parameters were further optimized.The optimal purification conditions were as follows:DM130macroporous resin was chosen,30 ℃was the optimal temperature for adsorption,the concentration of sample solution was 0.125 mg/mL,the volume of sample solution was 200 mL,adsorption flow rate was 6 mL/min,and eluted with 95% ethanol,elution flow rate was 4 mL/min.Under these conditions,atractylenolide I and atractylenolide II showed 32.3,and 25.6 fold increases in solid content,respectively.
