枸骨叶提取物对酪氨酸酶的抑制与抗氧化作用

摘要: 本实验以枸骨叶为研究对象,经75%乙醇浸提,用极性递增的溶剂分离制备获得枸骨叶提取物,测定了枸骨叶提取物对酪氨酸酶的抑制作用,并使用1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基清除的方法检测了枸骨叶提取物抗氧化能力。实验结果表明:枸骨叶三氯甲烷和正丁醇提取物对酪氨酸酶具有较强的抑制作用,IC50值分别达到0.414 mg/mL和0.667 mg/mL,均为混合型可逆抑制类型。同时,二者均具有一定的清除DPPH自由基的能力,IC50值分别为0.132 mg/mL和0.039 mg/mL,表明枸骨叶提取物同时具有抑制酪氨酸酶活性和抗氧化的功效,在美容、药物及食品等行业具有潜在的应用价值。

Abstract: In this experiment,the anti-tyrosinase and antioxidant activities of Ilex cornuta leaves extract (LIC) were measured.Ethanol extraction and increasing solvent polarity separation was used to obtain the component of LIC,its inhibition on tyrosinase and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical were measured.The result showed that the trichloromethane extract and butyl alcohol extract had strong inhibition on tyrosinase,the value of IC50 reached 0.414 and 0.667 mg/mL respectively.The kinesis study showed that their inhibition on tyrosinase were both reversible and inhibition types were both mixed.The trichloromethane extract of KI and KIS  were 0.269 mg/mL and 0.555 mg/mL;KI and KIS  of butyl alcohol extract were 0.273 mg/mL and 0.944 mg/mL.Meanwhile,both of them was found to have certain scavenging effect,IC50 was 0.132 and 0.039 mg/mL,which showed that LIC extract both had anti-tyrosinase and antioxidant efficacy,revealed it had potential application value in cosmetic,medicine and food,etc.
