一种蚯蚓体腔液蛋白的分离纯化及其生物活性

Purification of Earthworm Protein from Coelomic Fluid and Its Bioactivity

摘要:

赤子爱胜蚓体腔液含有多种蛋白并具有广泛的生物学活性,本研究采用6 V电压刺激蚯蚓分泌体腔液,通过离心、超滤分离得到蚯蚓体腔液蛋白粗品;然后将蛋白粗品经过凝胶层析和离子交换层析纯化,得到一种蚯蚓体腔液蛋白。以离子交换色谱分离到的蚯蚓体腔液蛋白样品为受试样品,紫外光谱扫描显示该蛋白在280 nm处有最大吸收峰,SDS-PAGE凝胶电泳测定其分子量约为38.6 KDa,比浊抑菌试验结果显示低浓度受试样品作用大肠杆菌1 h出现抑制效果,作用金黄色葡萄球菌3h出现抑制效果,表明该样品具有一定抑制细菌生长的活性,此外,该蛋白还具有较强的溶解鸡红细胞的作用,溶解红细胞的最低浓度为0.39 µg/mL,提示此蛋白的生物活性可能是通过破环细胞结构而实现的。

Abstract:

The coelomic fluid of earthworm Eisenia foetida exhibit extensively biological activity previously. Earthworm was induced by 6-Voltages electric stimulation to secrete coelomic fluid, followed by centrifugation and ultrafiltration to obtain a crude protein mixture. The crude extracts have been purified by means of gel chromatography and anion-exchange chromatography in sequence, as resulted in obtaining a kind of pure protein. Ultraviolet analysis indicated the protein absorption wavelength 280 nm, and SDS-PAGE determined the protein molecular weight 38.6 KDa. The further bioactive study showed that the protein displayed the inhibition effects on the growth of Escherichia coli in 1 h and Staphylococcus aureus in 3 h, as well as lysis effects on chicken red cells (minimum effect concentration 0.39 µg/mL). It suggested that activity of the protein could be come from by a means of damage-cell structure.
