大叶醉鱼草内生真菌LL3026菌株杀虫活性及ITS-5.8S rDNA序列分析

Insecticidal Activity and Sequences Analysis of ITS-5.8S rDNA of Endophyfic Fungi LL3026 from Buddleja davidii

摘要:

从大叶醉鱼草的叶子中分离得到一株内生真菌LL3026,以卤虫模型测稀释后发酵液的杀虫活性,结果表明LL3026发酵液杀虫活性较强,且温度、光照及紫外照射对LL3026发酵液杀虫活性影响不显著;采用分子生物学方法对LL3026 菌株rDNA的ITS基因(ITS-5.8S rDNA)进行PCR扩增、测序,构建系统发育树。ITS基因显示其属于刺盘孢属真菌。

Abstract:

An endophytic fungus LL3026 was isolated from leaves of Buddleja davidii. The insecticidal activity of the diluted fermentation broth was tested through the effect of anti-brine shrimp,and the results showed that the insecticidal activity was very strong. Revised mortality was remained 100% till LL3026 fermentation broth was diluted to 1/16,and LD50 was 55. There were no significant changes when temperature and the time of irradiation and UV were varied. The complete 5.8S rDNA sequence of LL3026 was amplified and analyzed through molecular biology methods,and phylogenetic tree was constructed. The analysis of DNA sequences showed that LL3026 strain was belonged to Colletotrichum sp..
