肾茶水提物喷干粉对D-半乳糖所致衰老小鼠学习记忆功能的影响

Effects of Orthosiphon stamineus Aqueous Extract on the Learning Memory  and Antioxidant Capacity of D-galactose Induced Aging Mice

摘要: 为观察肾茶水提物喷干粉(Orthosiphon stamineus aqueous extract,OSAE)的延缓衰老作用,采用D-半乳糖连续颈背部皮下注射诱导亚急性衰老小鼠模型,造模同时给予OSAE低、中、高剂量灌胃,连续4周。观察衰老小鼠在Moriss水迷宫检测学习记忆功能,测定脑组织中一氧化氮(NO)、超氧化物歧化酶(SOD)、单胺氧化酶(MAO)、三磷酸腺苷酶(Na+-K+-ATPase)、丙二醛(MDA)含量。结果显示,与模型组比较,OSAE高、中剂量组均能使小鼠逃避潜伏期明显缩短,原平台象限游泳时间明显延长(P<0.01或P<0.05);OSAE高剂量组SOD、NO、Na+-K+-ATPase活性显著升高(P<0.01或P<0.05),MDA、MAO水平显著下降(P<0.01或P<0.05)。表明OSAE可明显改善衰老小鼠学习记忆功能,提高抗氧化能力,从而可能具有较好的延缓衰老的作用。

Abstract: This study was conducted to investigate the anti-aging effect of Orthosiphon stamineus aqueous extract (OSAE).The subacute aging model mice were induced by injecting D-galactose subcutaneously.At the same time,low,medium and high dose of OSAE were administered intragastrically once daily for 4 weeks.Signs of aging were observed.The Moriss Water Maze test was used to evaluate the learning and memory function.The levels of NO,SOD,Na+-K+-ATPase,MAO and MDA in the serum and brain tissue were measured.The results showed that the high and middle dose of OSAE shortened the escape latency of the aging model mice,while prolonged the original platform quadrant swimming time significantly compared with the model group (P<0.01, P<0.05).High and middle dose of OSAE increased the SOD,NO,Na+-K+-ATPase activities of the aging model mice,and decreased the MAO and MDA content significantly (P<0.01, P<0.05).It indicated that OSAE can improve the learning and memory ability and increase antioxidant capacity of aging mice,which may has a good anti-aging effect.
