鼠尾草酸对Aβ损伤海马神经元的保护作用研究

Protective Effect of Carnosic Acid on β-Amyloid Protein Induced Hippocampal Neuron Injury

摘要: 采用Aβ诱导建立海马神经元损伤模型,CCK-8检测神经元的活性,RT-PCR检测神经元凋亡相关基因Caspase-3 mRNA的表达,探讨了鼠尾草酸对Aβ所致海马神经元损伤的保护作用及其机制。结果显示,浓度在5~10 μmol/L时,鼠尾草酸预处理能显著下调Aβ损伤导致的Caspase-3 mRNA表达的升高,增强神经元活力。表明鼠尾草酸预处理可以保护Aβ所致小鼠海马神经元的损伤,其机制可能与鼠尾草酸调控神经元凋亡相关基因caspase-3 mRNA的表达有关。

Abstract: The hippocampal neuron injury model established with β-amyloid protein (Aβ) was used to study the protective effects of carnosic acid on the toxic damage of hippocampal neuron.The viability of neuron was determined with CCK-8 assay,and mRNA levers of Caspase-3 were measured by RT-PCR in each experimental groups to analysis the influences of carnosic acid on the injured neuron.The results showed that the ability of carnosic acid protecting neuron from injury was enhanced obviously at 5 and 10 μmol/L,and the expression of Caspase-3 mRNA were decreased while the livability of neuron was increased.These results indicated that carnosic acid can protect Aβ induced hippocampal neuron injury,which was possibly through Caspase-3 signaling pathway.
