桑叶中聚戊烯醇结构与含量测定

摘要: 本文建立了高效液相色谱法确定桑叶聚戊烯醇的异戊烯基单元数和含量的方法,采用Thermo C18 ODS-2(250 mm×4.6 mm,5 μm)色谱柱,流动相为甲醇-异丙醇(8:7,v/v),流速为1.50 mL/min,柱温为25 ℃,检测器为PDA。测定结果表明,桑叶中聚戊烯醇的异戊烯基单元数为10~12,聚戊烯醇含量为0.731%。该方法准确度高、重现性好,能快速测定桑叶中聚戊烯醇的异戊烯基单元数和含量,为桑叶资源的开发提供理论依据。

Abstract: In this study,a high performance liquid chromatography (HPLC) method was developed to determine the number of isopentenyl units and the concentration of polyprenols in mulberry leaf.A Thermo C18 ODS-2 (250 mm × 4.6 mm,5 μm) column was used for chromatographic separation with methanol-isopropanol (8:7) as the mobile phases at a flow rate of 1.50 mL/min.The PDA detection wavelength was set at 210 nm,and the column oven temperature was set at 25 °C.The results showed that the number of isopentenyl units was 10 to 12 and the concentration of polyprenols in mulberry leaf was 0.731%.The developed HPLC method was accurate and reproducible.It can be used for the fast detection and determination of polyprenols in mulberry leaf.
