野马追的化学成分及其解热作用研究

摘要: 从野马追(Eupatorium lindleyanum DC.)中分离得到6个化合物,通过波谱数据分析,分别鉴定为:香草酸(1)、正二十八烷酸(2)、β-谷甾醇(3)、胡萝卜苷(4)、蒙花苷(5)、5,8,4′-三羟基-7,3′-二甲氧基黄酮(6),其中化合物2、5和6为首次从该植物中分离得到。采用UPLC/Q-TOF-MS法,分析野马追乙醇提取物的化学成分,并考察野马追对LPS诱导的大鼠发热模型的解热作用及其机制。腹腔注射脂多糖(100 μg/kg)法建立大鼠发热模型,记录灌胃给药后0.5、1、1.5、2、2.5、3、3.5、4、4.5、5、5.5、6、7、8 h大鼠的体温变化,绘制体温曲线;测定8 h大鼠肝组织髓过氧化酶活性和血清中肿瘤坏死因子-α和白介素-6含量。研究发现模型组大鼠体温呈明显三相热变化趋势,在造模后4 h出现升温峰值(1.97 ℃),8 h后体温恢复正常;野马追醇提物显著地降低了大鼠体温峰值;同时有效地抑制了肝组织中髓过氧化酶活性,降低了血清中肿瘤坏死因子-α、白介素-6含量。

Abstract: In this study,chemical composition of the ethanol extract of Eupatorium lindleyanum DC.was analyzed by UPLC/Q-TOF-MS.Six compounds were isolated from E.lindleyanum. These compounds were identified as vanillic acid (1),n-octacosanoic acid (2),β-sitosterol (3),daucosterol (4),linarin (5),5,8,4′-trihydroxy-7,3′-dimethoxy flavone (6). Compound 2,5 and 6 were obtained from this plant for the first time.In addition,the antipyretic effect and mechanism of E.lindleyanum on LPS-induced febrile response in rats was observed and elucidated.After the LPS challenge,the temperature of rats was recorded every 30 min,it lasted for 6 h,and then 7 h,8 h.Myeloperoxidase (MPO) activities in liver tissues was examined,the levels of inflammatory cytokine tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in serum were measured using ELISA.As a result,the rats of model group showed obvious three-phase fever and the heating peak value (1.97 ℃) appeared at 4 h.The ethanol extract of E.lindleyanum (EUP-EtOH) significantly decreased the body temperature of rats,and at 8 h following LPS challenge effectively inhibited MPO activity in liver tissues,TNF-α and IL-6 levels in serum.
