银杏叶聚戊烯醇急性和遗传性毒理学研究

Estimation of Acute and Genetic Toxicity of Polyprenols from Ginkgo biloaba L.

摘要: 本论文通过小鼠体内急性毒性试验、Ames试验、小鼠骨髓嗜多染红细胞微核试验和体外CHL细胞染色体畸变试验,对银杏叶聚戊烯醇急性和遗传性毒理进行评价。结果表明银杏叶聚戊烯醇属无毒级,最大给药量(21.5 g/kg)对小鼠增长、脏器系数和血液生理生化指标没有影响。Ames试验中,剂量达5000 μg/皿,在加与不加S9的条件下,TA97、TA98、TA100、TA102的自发回变菌落数均在正常范围内,对标准测试菌TA97、TA98、TA100和TA102均未检出明显的诱变活性(P>0.05)。微核和畸变试验表明,GBP剂量达10 g/kg b·wt,雌雄小鼠嗜多染红细胞/正染红细胞的比值与阴性对照组比较无统计学意义(P>0.05),对骨髓红细胞系统的增殖无明显受抑,对嗜多染红细胞微核的观察无明显影响。GBP各剂量组间小鼠染色体畸变的初级精母细胞率与溶剂对照值之间均无统计学差异(P>0.05),对雄性小鼠睾丸初级精母细胞染色体无诱变活性。因此,银杏叶聚戊烯醇无毒、无致癌、无致畸和无突变作用,为GBP保健食品和新药的开发提供安全理论依据。

Abstract: In this study,standardized acute toxicity test in mice and Ames test,bone marrow polychromatie erythrocyte micronucleus test and primary spermatocyte chromosome aberration tests in rats were conducted to evaluatre the acute and genetic toxicity of polyprenols of Ginkgo biloba L (GBP),a new type of lipids with isoprenyl units.The results showed that GBP was nontoxic,the maximum drug dose was 21.5 mg/kg in mice.No difference was detected between different groups on the body weight,growth organ/body weight ratio of rats and biochemical indexes (P>0.05).When GBP dose was 5000 μg/dish with or without S9,spontaneous revertant colonies of TA97,TA98,TA100 and TA102 were within the normal range,and no significant mutagenic activity were detected in Ames assay (P>0.05). Compared with negative control group in the micronucleus test,when GBP dose was 10 g/kg b·wt,the ratio of polychromatic erythrocytes to positive red cells in rats was not statistically significant (P>0.05),and proliferation of bone marrow erythrocytes system had no obvious inhibition,and the observation of polychromatie erythrocyte micronucleus had no obvious effect.Compared with negative control group in the chromosome aberration test,the aberrations rate of primary spermatocytes chromosome was no statistical difference (P>0.05),and no significant mutagenic activity of primary spermatocytes chromosome for male mouse were detected.Therefore,GBP was non-toxic,non-carcinogenic,non-teratogenic and non-mutagenic.
