摘要: 本研究采用70%乙醇超声提取和溶剂萃取获得槟榔提取物及其不同极性部位,采用200 μmol/L过氧化氢损伤SH-SY5Y细胞,建立体外模拟氧化应激损伤模型,采用MTT法检测细胞的存活率,评价槟榔提取物对过氧化氢损伤的SH-SY5Y细胞的保护作用。研究结果表明:除石油醚提取物对过氧化氢损伤后的SH-SY5Y不显示保护作用外,乙酸乙酯、正丁醇和水提取物均具有明显保护作用。其中乙酸乙酯部分分离得到的化学成分(2S,3R)-ent-catechin表现出较高的抗氧化活性,其体内生物活性和作用机制尚需进一步研究。

Abstract: Areca catechu nut extract and its different polar fractions were obtained using 70% ethanol extraction and solvent partition.The model of oxidative stress was established based on 200 μmol/L hydrogen peroxide injury of SH-SY5Y cell and cell viability was measured by MTT assay.Protective effects of all the samples on damaged SH-SY5Y cells induced by H2O2 were evaluated.The research results showed that ethyl acetate,n-butanol and water extracts have obvious protective effect except for petroleum ether part.Furthermore,(2S,3R)-ent-catechin isolated from EtOAc fraction showed higher antioxidant activity,its in vivo biological activity and mechanism need to be further studied.
