超声辅助双水相法提取葡萄籽中原花青素

Extraction of Procyanidins from Grape Seeds Using Ultrasonic-assisted Aqueous Two-phase System

摘要: 采用浊点法确定硫酸铵-乙醇-水和磷酸二氢钠-乙醇-水体系的相图,比较确定提取介质,并以提取率为指标,通过单因素试验和响应面法考察超声功率、超声时间、料液比对提取率的影响。结果表明,原花青素的最佳提取工艺条件为(NH4 )2SO4 23%(w/w),乙醇20%(w/w),超声功率750 W,超声时间44.5 min,料液比1:28 (g/g),提取温度25  ℃。在此条件下提取率为9.04 mg/g。原花青素对DPPH自由基有较强的清除能力,且超声辅助双水相提取葡萄籽中原花青素抗氧化能力明显高于一般回流提取。该工艺稳定可行,结果重现性较好。

Abstract: The phase diagrams of (NH4 )2SO4  or NaH2PO4+ethanol+water systems were developed by titration method.The effects of ultrasonic power,ultrasonic time and ratio of material to solution,on the extraction yield,were studied by single factor experiments and response surface methodology.The result showed that the optimal extraction conditions were (NH4 )2SO4  concentration of 23% (w/w),ethanol concentration of 20% (w/w),ultrasound input power of 750 W,ultrasound time of 44.5 min,ratio of solvent to solid of 28 (g/g) and extraction temperature of 25  ℃.The extraction yield was 9.04 mg/g under the optimal conditions.The Procyanidins has a strong ability of scavenging DPPH,and the antioxidant capacity of procyanidins extracted from grape seeds using ultrasonic-assisted aqueous two-phase system significantly higher than that of general reflux extraction.The technology were stable and feasible.
