荷包山桂花根化学成分研究

摘要: 研究荷包山桂花75%乙醇提取部分的化学成分。应用硅胶柱层析、Sephadex LH-20葡聚糖凝胶、制备高效液相等方法分离纯化化合物,并通过核磁共振、质谱等方法对化合物进行结构分析鉴定。从荷包山桂花根75%乙醇提取部分分离鉴定了10个化合物,分别为:3,6′ -二芥子酰基蔗糖(1)、顺式松柏苷(2)、α-菠甾醇(3)、2′ -脱氧胸苷(4)、α-菠甾醇-3-氧-β-D-吡喃葡萄糖苷(5)、β-谷甾醇(6)、丁香酸葡萄糖苷(7)、葡萄糖(8)、蔗糖(9)、对羟基苯甲醛(10)。化合物2~5和7为首次从荷包山桂花中分离得到。采用DPPH(1,1-二苯基-2-三硝基苯肼)和ABTS(2,2′ -连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸))自由基清除实验对化合物1~7、10进行了抗氧化活性研究,结果表明仅化合物1具有较强的清除自由基活性。

Abstract: To investigate the chemical constituents from the 75% alcohol fraction of the roots of Polygala arillata Buch.-Ham.ex D.Don.Column chromatographies of silica gel,preparative HPLC and Sephadex LH-20 were applied to isolate the chemical constituents.Pure compounds were elucidated by means of 1H NMR,13C NMR and LC-MS.Ten compounds were isolated and identified as β-D-(3-O-sinapoyl)-fructofuranosyl-α-D -(6-O-sinapoyl)-glucopyranoside (1),cis-coniferin (2),α-spinasterol (3),2′-deoxythymidine (4),α-spinasteryl 3-O-β-D-glucopyranoside (5),β-sitosterol (6),glucosyringic acid (7),glucose (8),sucrose (9),p-hydroxybenzaldehyde (10).Compounds 2-5 and 7 were obtainted from Polygala arillata Buch.-Ham.ex D.Don for the first time.The antioxidant activities of compounds 1~7 and 10 were tested by using the 2,2-diphenyl-1-picryhydrazyl (DPPH) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assays,and only compound 1 exhibited moderate antioxidant activity compared to ascorbic acid.
