Ebola假病毒的构建及其在中药抗病毒活性筛选方面的应用

Construction of Ebola Pseudovirus and Screening of Antiviral Activity of Traditional Chinese Medicine

摘要: 本实验利用细胞水平重组病毒技术,使病毒质粒和HIV质粒转染293T细胞构建Ebola、H5N1、Lassa假病毒,收集假病毒,于96孔板内感染A549细胞。利用Luciferase荧光底物检测感染效力。感染成功后进行抗病毒中药的筛选,筛选得到32个药品对Ebola假病毒有效。再通过毒性筛选排除药物毒性因素,初筛得到地龙、50%乙醇矮地茶提取物、75%乙醇矮地茶冷浸物等26个抗Ebola病毒中药。下一步可对其进行进一步提取纯化,寻找体外抗Ebola病毒的有效组分或成分。

Abstract: In this study,Ebola,H5N1 and Lassa Pseudoviruses were constructed from 293T cells transfected with virus plasmids and HIV plasmids by cell-level recombinant virus technology.Luciferase fluorescence substrate was used to detect infection efficacy.After screening,32 drugs were screened for Ebola virus.After eliminating the toxic factors through toxicity screening,26 anti-Ebola herbal medicines,such as earthworm,50% ethanol extract and 75% ethanol cold extract,were obtained.The next step is to further extract and purify Ebola,and to find the effective components or components of Ebola virus in vitro.
