苦杏仁酶转化人参皂苷Rg1和Rb1制备稀有人参皂苷

The Preparation of Rare Ginsenoside by Enzymolysis of Ginsenoside Rg1 and Rb1 with Emulsin

摘要: 本研究以人参皂苷Rg1和Rb1为原料,通过苦杏仁酶水解的方法,采用核磁共振、质谱鉴定酶解产物的结构,分别为人参皂苷Rh1、人参皂苷Rd、人参皂苷C-K。苦杏仁酶水解Rg1的途径为Rg1→Rh1,在室温下搅拌反应5天, Rg1几乎不发生反应;在温度为50 ℃条件下搅拌反应5天,得到唯一产物Rh1;苦杏仁酶水解Rb1的途径为Rb1→Rd→C-K,反应在室温下搅拌反应5天得到唯一产物Rd,在温度为50 ℃条件下搅拌反应5天,得到Rd与C-K的混合物。杏仁酶水解人参皂苷Rg1和Rb1反应条件温和,副产物少。

Abstract: In this paper,emulsin could convert ginsenoside Rg1 to the single pruduct of rare ginsenoside Rh1.The pathway of the transformation was Rg1 → Rh1.The reaction did not take place at room temperature for 5 days,while the reaction happened at 50 ℃ for 5 days and Rh1 was obtained.And emulsin could transform ginsenoside Rb1 to the mixture of Rd and rare ginsenoside C-K.The transformation pathway of ginsenoside Rb1 to C-K was Rb1→Rd→C-K.Rd was obtained after treated with emulsin at room temperature for 5 days,Rd and C-K was obtained while treated with emulsin at 50 ℃ for 5 days.The products were separated and purified by silica gel column chromatography.The structures of the hydrolyzates were identified with 13C NMR,1H NMR and MS spectra.The reaction conditions of emulsin transform ginsenoside Rg1 and Rb1 are mild and the byproducts are rare.
