不同醇沉部位双参多糖的单糖组成及抗氧化活性研究

Monosaccharide composition and antioxidant activity of crude polysaccharides from Triplostegia glandulifera with different alcohol precipitation

摘要: 研究不同醇沉部位对双参多糖的单糖组成及抗氧化活性的影响。双参经沸水提取、脱蛋白和大孔吸附树脂柱层析处理得水溶性成分,再经40%、60%和80%醇沉得粗多糖 TGP-1、TGP-2和TGP-3。采用PMP-HPLC方法对粗多糖的单糖组成进行分析,以粗多糖对DPPH、羟自由基和超氧阴离子的清除作用作为评价指标,评价它们的体外抗氧化活性。结果表明,TGP-1和TGP-2的单糖组成相同而摩尔比不同,它们均含Man-GluN-GalA-Glc-Gal-Ara,其中,TGP-1摩尔比为0.06∶0.03∶ 0.33∶3.90∶0.63∶0.41;TGP-2摩尔比为0.08∶0.06∶1.05∶15.71∶0.92∶0.49;TGP-3中含有GalA-Glc-Gal-Ara,其摩尔比为0.49∶5.65∶0.50∶0.35。抗氧化活性测试结果表明,TGP-3抗氧化活性最强,TGP-1抗氧化活性最弱。

Abstract: To study the effect of different alcohol precipitation on the composition and antioxidant activity of Triplostegia glandulifera polysaccharides.Water-soluble components was obtained by boiling water extraction,deproteinized,and macroporous resin column chromatography,then the crude polysaccharides of TGP-1,TGP-2 and TGP-3 were obtained by 40%,60% and 80% alcohol precipitation.PMP-HPLC methodology was used to analyze the monosaccharide compositions of TGP-(1-3).Finally,their antioxidant capacity was studied by scavenging of DPPH,hydroxyl and superoxide anion free radical method.The results showed that the polysaccharides TGP-1 and TGP-2 were composed of D-mannose, D-glucosamine hydrochloride,D-galacturonic acid,D-glucose,D-galactose and D-arabinose,the molar ratio of TGP-1 is about 0.06∶0.03∶0.33∶3.90∶0.63∶041,and the TGP-2 is about 0.08∶0.06∶1.05∶15.71∶0.92∶0.49. Moreover,TGP-3 were composed of D-galacturonic acid,D-glucose,D-galactose and D-arabinose and their molar ratio was 0.49∶5.65∶0.50∶0.35. Antioxidant capacity test results showed that the antioxidant capabilities of TGP-(1-3) were obviously different,among which TGP-3 had the strongest antioxidant capacity,and TGP-1 had the weakest antioxidant capacity.
