数据挖掘中药治疗癫痫药对“陈皮-半夏”及其网络药理学作用机制分析

Data mining Chinese herbal medicine for epilepsy drugs analysis of "Chenpi-Banxia" and its network pharmacological mechanism

摘要:

探讨中药治疗癫痫的方药使用规律,挖掘出常用药对陈皮-半夏,借助网络药理学技术预测陈皮-半夏中的化学成分所潜在的作用靶点,探讨其治疗癫痫多成分,多靶点,多通路的作用机制。以中国全文期刊数据库(CNKI)、维普、万方收集治疗癫痫的中药复方,运用Apriori算法建立起关联模型,挖掘出数据库中隐藏的药物配伍规律,利用TCMSP、OMIM、STRING及DAVID数据库分析药物疾病靶点间相互作用关系及相关通路。在所筛选出符合标准的131篇文献中,有226个方剂中共使用221种中药,得到置信度最高的药对陈皮-半夏,陈皮-半夏得到的18个目标化合物共检索出268个靶点基因,癫痫检索出的相关靶基因为886个,两者相关靶基因进行匹配得到共同靶基因41个,通过 Cytoscape 获取PPI网络中得到AKT1、MAPK3、FOS等具有重要地位的基因在陈皮-半夏治疗癫痫中具有重要意义。关键靶基因的GO和KEGG富集分析可得到陈皮-半夏配伍治疗癫痫的作用机制主要涉及对药物的反应、脂肪细胞分化的正调节、Ras蛋白信号转导、运动行为等生物功能,通过调节Serotonergic synapse、Kaposi sarcoma-associated herpesvirus infection、Hepatitis B等多条通路从而发挥治疗癫痫的作用。得到陈皮-半夏药对配伍治疗癫痫,并分析以陈皮-半夏配伍治疗癫痫的作用机理,为中医药治疗癫痫提供了新方法和新思路。

Abstract:

To explore the rule of application of traditional Chinese medicine in the treatment of epilepsy,and to explore the potential targets of the chemical constituents in the Chenpi and Banxia through the use of network pharmacology techniques to explore the potential components,multi-target and multi-channel mechanism of action of epilepsy.China’s full-text journal database (CNKI),VTTMS,VANFUN are used to collect Chinese herbal compound for treating epilepsy,and Apriori algorithm is used to establish a correlation model,so as to explore the drug compatibility rules hidden in the database and analyze drugs using TCMSP,OMIM,STRING and DAVID databases to analyze the interactions and related pathways between disease targets.Among the 131 papers that met the criteria,221 Chinese medicines were used in 226 prescriptions,of which Chenpi-Banxia was the highest confidence drug,a total of 268 targets were retrieved from 18 target compounds,a total of 886 target genes related to epilepsy were obtained and 41 targets were obtained by matching the relevant targets of the effective component of the Chenpi and Banxia.The common targets obtained by Cytoscape acquired the genes of AKT1,MAPK3,FOS and other genes in the PPI network.It is of great significance in the treatment of epilepsy in the Chenpi-Banxia.GO and KEGG enrichment analysis of key target genes can obtain the combination of Chenpi-Banxia was mainly involved in the response of cells to oxidative stress,the reaction to reactive oxygen species,the reaction of cells to cadmium ions,and other biological processes and regulated Serotonergic synapse,Kaposi sarcoma-associated herpesvirus infection,Hepatitis B and other pathways in the treatment of epilepsy.The mechanism of Chenpi-Banxia combination in the treatment of epilepsy was analyzed,which provided a new method and new idea for the treatment of epilepsy with traditional Chinese medicine.
