油橄榄叶甲醇提取物多酚成分、抗氧化及抑癌活性研究

Study on polyphenols,antioxidant and anticancer activities of methanol extract from olive leaves

摘要: 以油橄榄叶为材料,利用HPLC测定其甲醇提取物(EOL)中多酚成分,并通过秀丽线虫和Hela细胞探究其抗氧化及抗肿瘤活性。结果表明,EOL中含芦丁、木犀草素-4′-O-葡萄糖苷、芹菜素-7-O-葡萄糖苷、木犀草素和芹菜素5种主要多酚成分。EOL显著提高秀丽线虫抗氧化酶SOD、CAT和GSH-Px的活性,降低MDA水平,使其氧化应激存活率提升了72.52%。此外,EOL通过上调Caspase 9和Caspase 3活性抑制Hela细胞增殖,且肿瘤细胞抑制率呈现浓度和时间依赖型。因此,EOL具有开发为抗氧化和抑癌药物的潜力。

Abstract:

The methanol extract of olive leaves (EOL) was obtained by ultrasonic-assisted method.The polyphenols of EOL were determined by HPLC,and they antioxidant and anti-tumor activities were explored by Caenorhabditis elegans and Hela cells.The results showed that EOL contains 5 main polyphenol components including rutin,luteolin-4′-O-glucoside,apigenin-7-O-glucoside,luteolin and apigenin.EOL significantly increased the antioxidant enzyme activities of SOD,CAT and GSH-Px,decreased the level of MDA,and increased the survival rate of C. elegans under oxidative stress by 72.52%.In addition,HeLa cells were treated with different EOL concentrations to determine the inhibition rate of HeLa cells and the underlying anti-tumor mechanism of EOL was also explored.The results showed that EOL inhibited the proliferation of Hela cells by up-regulating the activities of Caspase 9 and Caspase 3,and the tumor cells inhibition rate was concentration and time-dependent.Therefore,EOL has the potential to be developed as an antioxidant and anticancer drugs.
