放线菌菌株H4-5发酵菌丝体的化学成分研究

Chemical constituents from the fermented mycelia of actinomycete strain H4-5

摘要:

对采集于近海红树林底泥的放线菌H4-5发酵菌丝体的次级代谢产物进行研究。综合运用多种色谱技术对放线菌H4-5发酵菌丝体的95%乙醇提取物进行分离纯化,通过波谱学方法并结合文献数据比对共鉴定了13个化合物的结构,分别为环-(L-脯氨酸-L-天冬酰胺)(1)、尿嘧啶(2)、尿苷(3)、2′-脱氧尿苷(4)、胸腺嘧啶核苷(5)、β-腺苷(6)、2′-脱氧腺苷(7)、豆甾-7,22-二烯-3β,5α,6α-三醇(8)、麦角甾醇(9)、N-(2-苯乙基)乙酰胺(10)、methyl 8-(3-octyloxiran-2-yl)octanoate(11)、tetrahydro-5-oxo-2-furancaboxylic acid ethyl ester(12)、环-(L-亮氨酸-L-缬氨酸)(13)。通过MTT法检测了这些化合物对三株人癌细胞的毒性并且通过LPS诱导的巨噬细胞NO释放实验检测了它们的抗炎活性,结果显示这13种化合物对供试的癌细胞没有明显的生长抑制作用,但化合物2显示出一定的抗炎活性。

Abstract:

The secondary metabolites of actinomycete H4-5 fermented mycelium collected from the sediments of offshore mangroves were studied.A variety of chromatographic techniques were used to separate and purify compounds from the 95% ethanol extract of actinomycete H4-5 fermentation mycelium.By spectroscopic methods combined with literature data comparison,13 compounds were isolated and identified as cyclo-(L-Pro-L-Asn) (1),uracil (2),uridine (3),2′-deoxyuridine (4),thymidine (5),β-adenosine (6),2′-deoxyadenosine (7),stigmasta-7,22-diene-3β,5α,6α-triol (8),ergosterol (9),N-(2-Phenethyl)acetamide (10),methyl 8-(3-octyloxiran-2-yl)octanoate (11),tetrahydro-5-oxo-2-furancaboxylic acid ethyl ester (12),cyclo-(L-Leu-L-Val) (13).The cytotoxicity of these compounds to three human cancer cell lines was tested by MTT method,and their anti-inflammatory activity was tested by LPS-induced macrophage NO release test.The results showed that these 13 compounds had no obvious effect on the tested cancer cell lines, but compound 2 shows certain anti-inflammatory activity.
