海洋来源真菌Penicillium janthinellum F12次级代谢产物研究

Study on secondary metabolites of fungus Penicillium janthinellum F12 from the South China Sea

摘要: 利用单菌多次级代谢产物(OSMAC)策略对采自南海北部-35 m深的沉积物海洋真菌Penicillium janthinellum F12进行化学成分研究。运用反复硅胶柱层析、葡聚糖凝胶(Sephadex LH-20)柱层析、反向中压柱层析(ODS)等色谱学方法对发酵产物进行化学分离,利用NMR、MS等波谱学技术并结合文献鉴定化合物结构,并对化合物进行了初步的抗植物病原菌活性测试。从菌株Penicillium janthinellum F12的液体发酵提取物中分离得到1个大环内酯类化合物brefeldin A(1),从其大米固体发酵提取物中,分离到4个生物碱类化合物paspalinine(2)、13-desoxypaxilline(3)、paspaline(4)和verruculogen(5)。在50 μg/mL质量浓度下,化合物1对荔枝炭疽病菌和链格胞菌的抑制率分别为40.5%和33.0%。研究结果表明,通过改变培养方式,能激活Penicillium janthinellum F12沉默基因的表达,产生完全不同类型的次生代谢产物。

Abstract:

Using one strain many compounds (OSMAC) strategy to study the chemical diversity of Penicillium janthinellum F12 collected from a sedimentary environment at a water depth of 35 m in the South China Sea.The compounds were purified by repeated silica gel column,Sephadex LH-20 and reversed medium pressure column chromatography.The structures of compounds were identified by spectroscopic analysis(NMR and MS) as well as comparison with published researches.The anti-phytopathogenic activities were assayed by plate method.A macrolide,brefeldin A (1),was isolated from the liquid fermentation extract of Penicillium janthinellum F12,and four alkaloids,paspalinine (2),13-desoxypaxilline (3),paspaline (4) and verruculogen (5),were isolated from the solid fermentation extract of Penicillium janthinellum F12.At the concentration of 50 μg/mL,compound 1 showed anti-phytopathogenic activities against Colletotrichum gloeosporioides and Alternaria alternata with the inhibition rates at 40.5% and 33.0%.By changing the culture conditions,the silenced genes of Penicillium janthinellum F12 could be activated and different types of secondary metabolites could be produced.
