黄毛榕中具有α-葡萄糖苷酶抑制活性化合物的研究

Study on the compounds with α-glucosidase inhibitory activity from leaves of Ficus esquiroliana

摘要:

为研究黄毛榕叶中具有降糖活性的成分,综合运用硅胶柱色谱、ODS柱色谱、Sephadex LH-20凝胶柱色谱以及半制备高效液相色谱法从黄毛榕95%的乙醇浸提物中共分离得到9个化合物,并通过核磁共振(NMR)、高分辨质谱(HR-ESI-MS)等技术鉴定了这9个化合物的结构,分别为N-阿魏烷基-L-脯氨酸甲酯(1)、N-反式-对香豆酰酪胺(2)、3′,4′,5′-三甲氧基肉桂醇(3)、lcariside H14)、(-)-dendrolactone(5)、3-羟基-1-(4-羟基-3,5-二甲氧基苯基)-1-丙酮(6)、反式阿魏酸(7)、4′-羟基-3′,5′-二甲氧基肉桂醇(8)和对羟基反式肉桂酸(9),化合物1为新化合物,化合物2~68首次从黄毛榕中分离得到。α-葡萄糖苷酶抑制活性测试结果表明得到的苯丙素类化合物1~68显示一定的α-葡萄糖苷酶抑制活性,IC50值范围为76.25~263.18 μmol/L。

Abstract:

This study aims to investigate the chemical constituents from the leaves Ficus equiroliana H.Lév,together with their α-glucosidase inhibitory activities.Nine compounds were isolated and purified from the 95% ethanol extract of the title plant by using systematic separation methods,including silica gel,ODS,Sephadex LH-20  column chromatography  and semi-preparative liquid chromatography.Their structures were identified as N-trans-ferulic-L-proline methyl ester (1),N-p-coumaroyltyramine (2),3′,4′,5′-trimethoxycinnamyl alcohol (3),lcariside H1 (4),(-)-dendrolactone (5),3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone (6),trans-ferulic acid (7),sinapyl alcohol (8) and (E)-p-coumaric acid (9) by NMR and HR-ESI-MS.Compound 1 is a new compound,compounds 2-6 and 8 were isolated for the first time from the leaves of F. equiroliana.Compounds 1-6 and 8 showed α-glucosidase inhibitory activities with IC50 values of 76.25-263.18 μmol/L.
