不同来源段木灵芝子实体指纹图谱及灵芝酸A、灵芝酸B含量测定研究

Study on fingerprints of Ganoderma lucidum from different wood segments and content determination of ganoderic acid A and ganoderic acid B

摘要:

人工栽培灵芝常选用不同的段木进行,而其中成分存在较大差异,从而影响灵芝。为比较成分差异,建立灵芝子实体HPLC指纹图谱,测定灵芝酸A、B含量,为不同段木灵芝子实体的质量评价及栽培技术提供参考。采用Diamonsil C18(2)(250 mm×4.6 mm,5 μm),以乙腈-0.1%磷酸水溶液为流动相梯度洗脱,共标定了18个共有峰,指认了灵芝酸A、灵芝酸B。15批次灵芝子实体图谱相似度范围在0.601~0.986,主成分分析将其分为4类,正交偏最小二乘法-判别式分析(OPLS-DA)共筛选出7个VIP>1.0的差异性成分。经含量测定,发现柳树+桦树该段木中灵芝酸A含量为2.29 mg/g,灵芝酸B含量为0.47 mg/g,二者均为最高,推测其含量可能与栽培段木有关。该方法可用于评估不同来源段木灵芝子实体质量,筛选合适的灵芝栽培段木,指导灵芝栽培。

Abstract:

In the artificial cultivation of Ganoderma lucidum,different wood segments are often used and there are significant differences in their composition that affect G .lucidum.In order to compare the differences in components,HPLC fingerprints of G .lucidum was performed and the content of ganoderic acid A and B was determined,which provides a reference for the quality evaluation and cultivation techniques of different wood segments of G .lucidum.The analysis was performed on Diamonsil C18 (2) (250 mm× 4.6 mm,5 μm) with acetonitrile-0.1% phosphoric acid solution as the mobile phase in a gradient elution mode,a total of 18 common peaks were calibrated and ganoderic acid A and ganoderic acid B were identified.The similarity of the 15 batches of G .lucidum ranged from 0.601 to 0.986.Principal component analysis divides them into four categories,and orthogonal partial least squares discriminant analysis (OPLS-DA) selects seven differential components with VIP>1.0.The content determination showed that the content of ganoderic acid A and ganoderic acid B in the willow+birch wood segment was 2.29 mg/g and 0.47 mg/g,respectively.Both were the highest,suggesting that their content might be related to the cultivated segment of the wood.This method can be used to evaluate the quality of G .lucidum from different wood segments,screen suitable G .lucidum cultivated wood segments,and guide G .lucidum cultivation.
