Determination of Tripterine in Tripterygium hypoglaucum by Ionic Liquidbased Ultrasoundassisted Extraction Combined with RPHPLC

摘要: 本研究建立了以6种离子液体为萃取剂,超声辅助萃取结合高效液相色谱法测定火把花(昆明山海棠)中雷公藤红素含量的方法。采用Inertsil ODS4柱(3 μm,4.6×250 mm);以乙腈0.1%磷酸溶液(80∶〖KG-*2〗20)为流动相;检测波长:425 nm;柱温:30 ℃;流速:0.75 mL/min。结果显示,提取雷公藤红素的最佳萃取条件为0.6 mol/L [BMIm]PF6甲醇溶液,固液比为1∶〖KG-*2〗4(g/mL),在0.015~0.45 μg范围内呈良好的线性关系(r=0.999 9),平均回收率为99.25%,RSD为2.22%。实验表明该方法操作简便,准确可靠,稳定性好,且绿色环保,为火把花的质量控制提供了一种可靠的分析方法。

Abstract: Using six kinds of ionic liquids as extractants,a method of ultrasonicassisted extraction combined with HPLC was developed for the simultaneous determination of tripterine in Tripterygium hypoglaucum.The separation was performed on an Inertsil ODS4 column with the mobile phase of acetonitrile0.1% phosphoric acid (80:20),detection wavelength was 425 nm and column temperature was 30 ℃.Under the optimal extractions,the results showed that tripterine had the highest extraction yield through using 0.6 mol·L1 [BMIm]PF6 methanol solution as extraction solvent with the solidliquid ratio of 1:4 (g·mL1).The calibration curves showed a good linearity in the range of 0.015045 μg (r=0.999 9),with the average recovery of 99.25% and RSD is 2.22%.These results indicated that this method is convenient,accurate,reliable and environment friendly,providing a reliable analytical method for the quality control of Tripterygium hypoglaucum.
