藜麦种子总黄酮提取和含量测定方法的比较

Comparison on the Extraction and Determination Method of Total Flavonoids in Quinoa Seed

摘要: 利用响应面分析法比较了藜麦种子总黄酮含量提取的氯化铝显色法和硝酸铝显色法及其主要影响因素,以期获得藜麦种子最佳的黄酮提取方案。在单因素实验基础上,选取乙醇浓度、提取温度和提取时间为影响因素,以藜麦种子总黄酮得率为响应值进行响应面分析。氯化铝显色法的最佳提取条件是在乙醇浓度为7197%,提取温度为62.45 ℃条件下提取1.11 h,此时藜麦黄酮得率为5.322%,误差为7.68%;硝酸铝显色法的最佳提取条件是在乙醇浓度为84.43%,提取温度为61.13 ℃条件下提取1.24 h时,此时藜麦黄酮得率为8799%,误差为1.24%。硝酸铝显色法误差小,稳定性高,所得总黄酮含量较高,适合于藜麦种子总黄酮含量的提取测定。

Abstract: AlCl3 colorimetry method and Al(NO3)3 colorimetry method of total flavonoids from quinoa were studied by response surface methodology,in order to obtain the best extraction method.Based on single factor experiments,take ethanol concentration,extraction temperature and extraction time as main variables.With the total flavonoids yield,the content determination was optimized by response surface analysis method.The best extraction process for AlCl3 colorimetry method was to extract 1.1h in 62.45 ℃ water bath and 71.97% ethanol concentration,the yield of flavonoids was 5322% while error was 7.68%.The best extraction process for Al(NO3)3 colorimetry method was to extract 1.24 h in 61.13 ℃ water bath and 84.43% ethanol concentration,the yield of flavonoids was 8.799% while error was 1.24%.Compared with AlCl3 colorimetry method,Al(NO3)3 colorimetry method was efficient and stable for the determination of flavone in quinoa.
