小单孢菌FIM060152中抗肿瘤活性成分的提取分离纯化

摘要: 本文是探讨分离和提取纯化小单孢菌FIM060152发酵液中的抗肿瘤抗生素的方法。实验采用HZ816大孔吸附树脂柱层析、萃取、硅胶柱层析、半制备色谱等方法对菌株FIM060152的发酵液中次级代谢产物进行提取分离纯化,获得化合物FIM060152-C1。理化性质和UV、MS以及NMR波谱分析结果表明化合物FIM060152-C1为CalicheamicinγI1。化合物FIM060152-C1的抗肿瘤活性通过DNA断裂法进行检测,其断裂质粒pBR的最低浓度约为0.20 μg/mL。

Abstract: The purpose of the present study was to isolate,purify and identify antitumor compounds from the fermentation broth of Micromonospora FIM060152.The secondary metabolite was isolated and purified by macroporous resin column chromatography,silica gel chromatography and preparative chromatography;the structure was identified by physicochemical properties,LC-MS and NMR spectral data.The antitumor activity was detected by DNA damage.As a result,compound FIM060152-C1 was isolated from the strain and identified as CalicheamicinγI1.DNA damage exhibited that FIM060152-C1 had strong antitumor activity on plasmid pBR322,with MIC value of 0.2 μg/mL.
