二烯丙基二硫对IL-1β诱导SD大鼠关节软骨细胞的影响

Effects of Diallyl Disulfide on SD Rat Articular  Cartilage Cell Induced by IL-1β

摘要: 观察二烯丙基二硫(DADS)对IL-1β诱导SD大鼠关节软骨细胞的影响,除空白对照组外,其余各组加入IL-1β(10 ng/mL)诱导24 h后换DADS处理,通过CCK8实验、平板克隆实验检测增殖能力,Western blot实验检测MMP13、TIMP1和Collagen Ⅱ蛋白表达水平。结果发现:12.5和25 μmol/L DADS对IL-1β诱导SD大鼠关节软骨细胞有增殖作用(P<0.05),50和100 μmol/L DADS对IL-1β诱导SD大鼠关节软骨细胞有抑制作用(P<0.05);12.5和25 μmol/L DADS显著增加IL-1β诱导SD大鼠关节软骨细胞克隆集落形成能力(P<0.05),50 μmol/L DADS显著抑制IL-1β诱导SD大鼠关节软骨细胞克隆集落形成能力(P<0.05);DADS能显著抑制MMP13的表达,增加TIMP1和Collagen Ⅱ的表达。以上结果提示DADS对IL-1β诱导SD大鼠关节软骨细胞具有双向调节作用,25 μmol/L DADS保护关节软骨细胞的分子机制可能与DADS显著抑制MMP13的表达,增加TIMP1和Collagen Ⅱ的表达相关。

Abstract: To explore the effect of diallyl disulfide (DADS) on articular cartilage cell induced by IL-1β  in SD rats.The growth inhibition of articular cartilage cell induced by IL-1β in SD rats following DADS treatment was measured by CCK8 assay and plate colony formation assay; The protein expression of MMP13,TIMP1 and Collagen II were determined using Western blot.DADS (12.5 and 25 μmol/L) significantly increased the cell proliferation and high colony formation capacity; DADS (50 and 100 μmol/L) significantly suppressed the cell proliferation and lowered colony formation capacity (P<0.05); Western blot demonstrated DADS markedly down regulated the expression of MMP13,and increased the expression of Collagen II and TIMP1 in the cells.These results indicated that DADS had two-way regulation effect on SD rat articular cartilage cell induced by IL-1β, DADS (25 μmol/L) had a protective effect on SD rat articular cartilage cell induced by IL-1β, DADS (50 and 100 μmol/L) can inhibit articular cartilage cell by IL-1β in SD rats,the protection molecular mechanism was possibly related to downregulation of the expression of MMP13 by DADS,and increased the expression of Collagen II and TIMP1 on SD rat articular cartilage cell with osteoarthritis.
