不同品系丹参毛状根的诱导效应比较研究

Study on Inductive Effect of Salvia miltiorrhiza Hairy Root with Different Strains

摘要: 不同遗传背景丹参材料的质量存在差异,但其毛状根的诱导效应是否存在差异尚未验证。本试验利用发根农杆菌C58C1诱导7个不同遗传背景的丹参无菌苗产生毛状根,并比较其毛状根的诱导率、生长速率、有效成分含量等差异表现,结果表明,不同遗传背景的丹参材料,其毛状根诱导效应存在差异,其中Ds2品系的诱导率最低,显著低于其他6个品系;而Ds5、Ds6、Ds7 三个品系的生长速率显著低于Ds1、Ds3、Ds4;比较Ds1、Ds3、Ds4三个品系的有效成分含量,则发现其差异明显,且各有优势,首次验证了不同遗传背景丹参毛状根的诱导效应存在显著差异。同时,通过综合评价,筛选出三3个适宜毛状根高效发酵生产的优良丹参品系Ds1、Ds3、Ds4,其中生产迷迭香酸选用Ds1、Ds4,生产丹酚酸B选用Ds4,生产丹参酮Ⅰ选用Ds3,生产丹参酮ⅡA选用Ds1,为丹参毛状根技术的生产应用提供了实验依据。

Abstract: The quality of Salvia miltiorrhiza is different in different genetic backgrounds;however,the induction effect of Salvia miltiorrhiza hairy root has not been verified.In this experiment,transformed hairy roots of seven different Salvia miltiorrhiza strains were obtained by the infection of  Agrobacterium rhizogenes C58C1 and a hairy root culture system in vitro was established.By comparing induction efficiency,growth rate,content of active component from the above seven different genetic background Salvia miltiorrhiza's hairy root,the results showed that the induction effects of hairy root in different genetic backgrounds were different.Among them,the induction rate of Ds2 strain was lowest,which was significantly lower than that of the other 6 lines;the growth rates of three strains of Ds5,Ds6 and Ds7 were lower than Ds1,Ds3 and Ds4 strains;comparing the effective components content of three strains of Ds1,Ds3 and Ds4,it was found that the differences were obvious,and each had its advantages.Therefore,it was the first time to verify the induction effect of hairy root of Salvia miltiorrhiza in different genetic backgrounds.At the same time,through comprehensive evaluation,we screened three fine Salvia miltiorrhiza strains Ds1,Ds3 and Ds4 which were suitable for efficient fermentation of hairy roots.Rosmarinic acid was produced by Ds1 and Ds4,salvianolic acid B was produced by Ds4,and Ds3 was used to produce tanshinoneⅠ,Ds3 was used to produce tanshinoneⅡA.
