单羽火筒树化学成分研究

摘要: 采用色谱柱层析及半制备高效液相色谱的分离方法,以及质谱和核磁共振波谱的鉴定方法,从云南省德宏市民间利用的抗肿瘤药用植物单羽火筒树(九节狸)全株中分离、鉴定出了11个化合物,即山柰酚(1)、山萘酚-3-O-α-L-吡喃鼠李糖苷(2)、槲皮素(3)、异槲皮苷(4)、槲皮素-3-O-α-L-吡喃鼠李糖苷(5)、杨梅素(6)、杨梅素-3-O-α-L-吡喃鼠李糖苷(7)、Europetin(8)、Europetin 3-O-α-L-rhamnopyranoside(9)、(-)-儿茶素(10)和山楂酸(11)。采用MTS法测试了乙酸乙酯部位及11个化合物对5种肿瘤细胞株的体外细胞毒活性,结果表明单羽火筒树乙酸乙酯部位对人白血病HL-60细胞(IC50 =21.76 μg/mL)、人肝癌SMMC-7721细胞(IC50=17.04 μg/mL)和人肺癌A-549细胞株(IC50=18.00 μg/mL)的生长有抑制活性,从乙酸乙酯部位分离出来的化合物1对肝癌细胞株(SMMC-7721)的生长有抑制活性(IC50=27.61 μg/mL)。

Abstract: Through column chromatography and semi-preparative high performance liquid chromatography (HPLC),11 compounds were isolated from the whole plants of Leea asiatica with the common name of Jiu-Jie-Li,a traditional antitumor ethnomedicine used by local people in Dehong of Yunnan Province.These compounds were identified as kaempferol (1),kaempferol-3-O-α-L-rhamnopyranoside (2),quercetin (3),isoquercitrin (4),quercitrin (5),myricetin (6),myricetin-3-O-α-L-rhamnopyranoside (7),europetin (8),europetin 3-O-α-L-rhamnopyranoside (9),ent-catechin (10),and maslinic acid (11) by MS and NMR spectra.The cytotoxic activity against 5 tumor cell lines of the EtOAc-soluble part and 11 compounds were evaluated by an MTS assay.The result showed that the EtOAc-soluble part exhibited growth inhibitory activity against human leukemia HL-60 (IC50=21.76 μg/mL) cell line,human lung cancer A-549 (IC50=18.00 μg/mL) cell line,and human liver cancer SMMC-7721 (IC50=17.04 μg/mL) cell line,and kaempferol (1) from the the EtOAc-soluble part was active against SMMC-7721 cells with an IC50 value of 27.61 μg/mL.
