芳香植物假荜拔茎叶的化学成分及其胆碱酯酶抑制活性研究

Chemical Constituents from the Stem and Leaf of Piper retrofractum and Their Anti-cholinesterase Activity

摘要: 采用凝胶树脂(Toyopearl HW-40c)、正相硅胶、反相硅胶(ODS)、大孔树脂(MCI-gel CHP20P)等柱层析和半制备HPLC分离方法从胡椒科胡椒属植物假荜拔(Piper retrofractum)茎叶的70%丙酮提取物中分离得到10个化合物,包括1个生物碱衍生物(1)、6个酰胺类衍生物(2~7)、2个酚性成分(8和9)以及1个紫罗兰酮类衍生物(10)。运用波谱光谱学方法分别鉴定为:4-[2-甲酸基-5-(甲氧基甲基)-1H-吡咯-1-基]丁酸(1)、3-氯-4-羟基-2-哌啶酮(2)、反式-锯齿菊除虫素(3)、1-肉桂酰基哌啶(4)、(2E,4E)-胡椒碱(5)、胡椒新碱(6)、3-苯丙酰胺(7)、(2R/2S)-异奥卡宁7-O-β-D-吡喃葡萄糖苷(8a和8b)、香草酸(9)和布卢姆醇A(10),其中化合物1,7,8和10均首次从胡椒属中分离得到。采用Ellman比色法对部分化合物的胆碱酯酶抑制活性进行测试,活性测试结果表明化合物4对乙酰胆碱酯酶和丁酰胆碱酯酶均显示抑制活性,半数抑制浓度(IC50)分别为37.44±0.62 μM和3.03±0.11 μM;化合物5和6选择性抑制丁酰胆碱酯酶,半数抑制浓度(IC50)分别为49.43±2.13和24.70±0.90 μM。但是,其余化合物在100 μM浓度下均没有显示胆碱酯酶抑制活性。

Abstract: Ten compounds were isolated from the stem and leave of Piper retrofractum by using the method of Toyopearl HW-40c,silica gel,ODS,MCI-gel CHP20P and pre-HPLC,including one alkaloid (1),six amides (2-7),two phenols (8 and 9),and one ionone derivative (10).The structure elucidation was based on spectroscopic analysis as 4-[2-formyl-5-(methoxymethyl)-1H-pyrrol-1-yl]butanoic acid (1),3-chloro-4-hydroxy-2-piperidone (2),trans-pellitorin (3),1-cinnamoylpiperidine (4),(2E,4E)-piperine (5),piperanine (6),3-phenylpropanamide (7),(2R/2S)-isookanin-7-O-β-D-glucopyranoside(8a and 8b),vanillic acid (9),and blumenol A (10).Among them,compound 1,7,8 and 10 are reported from Piper plants for the first time.Some isolates were tested for their effect on anti-cholinesterase activity by Ellmans method.Compound 4 showed anti-acetylcholinesterase and anti-butyrocholinesterase activities with IC50 values of 37.44±0.62 μM and 3.03±0.11 μM,respectively.Compounds 5 and 6 showed selectively antibutyrocholinesterase activity with IC50 values of 49.43±2.13 and 24.70±0.90 μM,respectively.
