大黄素在行为模型上的抗抑郁作用

Anti-depressant Effects of Emodin on Animal Model

摘要: 为探讨大黄素(Emodin)的抗抑郁作用及其机制,选取雄性昆明小鼠随机分为空白对照组,盐酸氟西汀组(5 mg/kg)和大黄素组(200,100,50 mg/kg),连续灌胃给药7 d后,进行小鼠悬尾试验(TST)和小鼠强迫性游泳试验(FST);除空白对照组外,各组连续灌胃给药7d后,分别腹腔注射利血平(1 mg/kg),观察大黄素对利血平所致小鼠眼睑下垂和体温下降的拮抗作用。实验结果显示:在小鼠悬尾试验和小鼠强迫性游泳试验中,大黄素高剂量(200 mg/kg)能明显缩短小鼠在悬尾实验和强迫游泳实验中的不动时间(P<0.05)。在利血平诱导小鼠抑郁试验中,大黄素高剂量(200 mg/kg)能显著对抗利血平诱导的小鼠体温下降和眼睑下垂(P<0.05)。由此可见,大黄素具有明显的抗抑郁作用,其作用机制可能与单胺递质有关。

Abstract: To investigate the antidepressant effect and the possible mechanism of emodin in animal model.Mail Kunming mice were randomly divided into 5 groups:Control group (normal saline),fluoxetine group (5 mg/kg),emodin groups (200,100,50 mg/kg).After oral administration for 7 days,the tail-suspending test (TST),the forced swimming test (FST) and reserpine-antagonism test were carried out to evaluate the antidepressant effects of emodin.In particular,reserpine (1 mg/kg) was given by intrapertioneal injection before the reserpine-antagonism test.Compared with the control group,the immobility time of TST and FST were shorted in emodin high dose group (P<0.05).Compared with the model group,emodin high dose could significantly reverse the ptosis,and decrease body temperature induced by reserpine in mice (P<0.05).The results showed that emodin possesses significant antidepressant effect,and the mechanism may be related to monoamine neurotransmitters.
