青海省不同产地蒙古黄芪芒柄花苷和毛蕊异黄酮含量比较

Comparison of the content of ononinand and calycosin of Astragalus membranaceus in different producing areas in Qinghai province

摘要: 采用HPLC法测定青海省不同产地蒙古黄芪芒柄花苷和毛蕊异黄酮的含量,及土壤与生长指标和有效成分含量的相关性研究,为青海省不同产地蒙古黄芪药材品质综合评价提供参考资料及产区规范化栽培提供科学依据。采用Agilent HC-C18(250×4.6 mm,5 μm)色谱柱,流动相为乙腈和0.01%磷酸水,梯度洗脱,流速为1 mL/min,柱温30 ℃,检测波长260 nm。湟中县黄芪生长指标整体较好,而乐都县黄芪生长指标较差,以产量为指标,湟中县>互助县>民和县>湟源县>大通县>乐都县。不同产地的蒙古黄芪,其芒柄花苷和毛蕊异黄酮的含量差异较大。六个县中,芒柄花苷含量依次为大通县>乐都县>民和县>互助县>湟源县>湟中县;毛蕊异黄酮含量依次为互助县>大通县>乐都县>湟中县>民和县>湟源县。以芒柄花苷含量为采收指标,大通县为适宜产地,以毛蕊异黄酮为采收指标,互助县为适宜产地,综合两个含量指标及系统聚类分析结果,乐都县为最佳采收地。但是以外观品相和产量为采收指标,湟源县为最佳采收地。青海省19个产地芒柄花苷含量不低于0.294 mg/g,建议将蒙古黄芪含芒柄花苷不得少于0.02%定为青海省完善蒙古黄芪质量评价指标之一。

Abstract: Astragalus membranaceus from different producing areas in Qinghai province,and the correlation between soil and growth index and the content of effective components was studied,so as to provide references for the comprehensive quality evaluation of Astragalus membranaceus from different producing areas in Qinghai province and scientific basis for the standardized cultivation in producing areas.Agilent HC-C18 (250 × 4.6 mm,5μm) column was used.The mobile phase was acetonitrile and 0.01% phosphoric acid water with gradient elution.The flow rate was 1 mL/min,the column temperature was 30 ℃,and the detection wavelength was 260 nm.The growth index of Huangzhong County is better,but that of Ledu County is worse.Taking yield as index,Huangzhong County > Huzhu County > Minhe County > Huangyuan County > Datong County > Ledu County.Astragalus mongolica from different habitats has different contents of anthocyanin and isoflavone of stamen.Among the six counties,the content of mangiferin was Datong County > Ledu County > Minhe County > Huzhu County > Huangyuan County > Huangzhong County;the content of stamen isoflavone was Huzhu County > Datong County > Ledu County > Huangzhong County > Minhe County > Huangyuan County.Taking the content of Ononoside as the harvesting index,Datong County as the suitable harvesting area,mulberry isoflavone was taken as harvesting index and Huzhu County as suitable producing area.Ledu County as the best harvesting area by synthesizing two content indexes and cluster analysis.However,Huangyuan County is the best harvesting area with appearance and yield as harvesting indicators.The content of Ononoside in 19 producing areas of Qinghai Province is not less than 0.294 mg/g.It is suggested that the content of ononoside in Astragalus mongolica should not be less than 0.02% as one of the quality evaluation indexes for improving the quality of Astragalus mongolica in Qinghai Province.
