植酸抗结肠癌细胞的作用及其机制

摘要: 为研究植酸(IP6)抑制结肠癌细胞增殖的作用及其机制,我们采用MTT、克隆形成及TUNEL染色检测植酸对结肠癌细胞增殖及凋亡的影响;免疫印迹(Western blot)法检测细胞凋亡相关标志蛋白cleaved-caspase3、PARP、Cyt C、Bax;自噬相关标志蛋白Beclin1、LC3、p62的表达以及自噬AKT-mTOR通路的抑制情况。结果显示植酸对结肠癌细胞HCT116具有明显的抑制作用且呈药物浓度依耐性;并且植酸通过抑制AKT-mTOR通路诱导结肠癌自噬而不影响细胞凋亡而达到抑制结肠癌增殖的效果。因此植酸通过抑制AKT-mTOR信号通路来激活细胞自噬,从而抑制结肠癌细胞的增殖。

Abstract: To investigate the effect and mechanism of phytic acid (IP6) on the proliferation of colorectal cancer cells,we performed MTT,clone formation and TUNEL staining to detect the effect of phytic acid on proliferation and apoptosis of colorectal cancer cells;Western blot was detected the expression of apoptosis-related proteins caspase3,PARP,Cyt C,and Bax;the autophagy-related proteins Beclin1,LC3,p62 and AKT-mTOR pathway.The results showed that phytic acid had a significant inhibitory effect with drug-dose-dependent on colorectal cancer cell HCT116;and phytic acid can inhibit colorectal cancer proliferation by inhibiting AKT-mTOR pathway to induce autophagy without apoptosis.Therefore,phytic acid activates autophagy by inhibiting the AKT-mTOR signaling pathway to inhibit the proliferation of colorectal cancer cells.
