小分子果胶对Ⅱ型糖尿病模型小鼠的治疗作用及机制研究

Effect and mechanism of low-molecular-weight citrus pectin on type II diabetic mice

摘要: 为观察II型糖尿病模型小鼠连续10周经口灌胃给予小分子果胶对小鼠血糖血脂的影响并初步探讨其作用机制。实验选用db/db小鼠,随机分为II型糖尿病模型对照组和3个剂量给药组,以db/m小鼠为正常对照组,给药组分别连续10周经口灌胃给予等体积的30、20、13.3 mL/kg的小分子果胶,正常对照组和模型对照组动物给予等量生理盐水。观察小分子果胶对II型糖尿病小鼠体重、血糖血脂、胰岛素及葡萄糖耐量的影响。研究发现II型糖尿病模型小鼠连续10周经口灌胃给予小分子果胶后,小鼠血糖、血清甘油三酯、胆固醇含量显著降低,且小鼠葡萄糖耐受能力显著提高;但小鼠体重、血清低密度脂蛋白、高密度脂蛋白以及胰岛素无显著影响,小鼠胰岛素耐受能力也无显著影响。研究结果提示,小分子果胶可调节体内糖、脂等代谢过程,对II型糖尿病具有一定的治疗作用。

Abstract: This study was to investigate hypoglycemic effect of low-molecular-weight citrus pectin (LCP) in type II diabetic mice by oral gavage for 10 weeks and to explore its mechanism.The db/db mice were divided into the type II diabetes model group and three dose administration groups,as well as the db/m mice were used as the normal control.The administration groups were given an equal volume of 30,20,13.3 mL/kg LCP by oral gavage for 10 weeks,and the control and model group were given the same volume of saline.The body weight,blood glucose,lipids,insulin and glucose tolerance of the mice were measured,and then the effects of LCP on these indicators in type II diabetic mice were analyzed.The blood sugar,serum triglyceride and cholesterol levels were significantly decreased,and the glucose tolerance of mice was significantly increased in the mice with type II diabetes were given LCP for 10 weeks.There was no significant effect on serum low-density lipoprotein,high-density lipoprotein,insulin and insulin tolerance in mice.The results suggest that LCP can regulate the metabolism of blood sugar and lipids and has a potential therapeutic effect on type II diabetes.
