龙血竭的化学成分及降血糖活性研究

摘要:

对龙血竭乙醇提取物的正丁醇萃取物的化学成分及降糖活性进行研究。通过硅胶、Sephadex LH-20、Rp-C18和高效液相色谱反复层析纯化,从龙血竭乙醇提取物的正丁醇萃取物中分离得到6个化合物,运用现代波谱技术将它们的结构分别鉴定为3-methyl resveratrol(1)、dracaenin A(2)、7-hydroxy-3-(4′-methoxybenzyl)-4-chromanone(3)、diosgenin-3-O-α-L-pyranorhamnosyl(1→2)-β-D-glucopyranoside(4)、(22E)-3β-acetoxystigmasta-5,22-diene(5)和nonadecyl alcohol(6)。其中,化合物4为首次从龙血竭中分离得到。对龙血竭乙醇提取物的正丁醇萃取物各分段部位和单体化合物进行α-葡萄糖苷酶抑制活性检测,发现化合物1和2具有明显的抑制活性,其IC50值分别为14.62±0.083和16.03±0.076 μM,且化合物2的α-葡萄糖苷酶抑制活性为首次报道。

Abstract:

To investigate the chemical components of Resina Draconis and evaluate their hypoglycemic activities,six compounds,3-methyl resveratrol (1),dracaenin A (2),7-hydroxy-3-(4′-methoxybenzyl)-4-chromanone (3),diosgenin-3-O-α-L-pyranorhamnosyl(1→2)-β-D-glucopyranoside (4),(22E)-3β-acetoxystigmasta-5,22-diene (5) and nonadecyl alcohol (6) were isolated from the n-butanol fraction of Resina Draconis by chromatographic techniques including column chromatography of silica gel,Sephadex LH-20,Rp-C18 and HPLC.Their structures were mainly elucidated by NMR and MS spectroscopic techniques.Compound 4 was isolated from the Resina Draconis for the first time.Four subfractions of the n-butanol fraction of Resina Draconis and 6 compounds were tested for their inhibitory activities against α-glucosidase,compounds 1 and 2 exhibited potent inhibitory activities with IC50 values of 14.62 ± 0.083 and 16.03 ± 0.076 μM,respectively.The inhibitory activity of compound 2 on α-glucosidase was reported for the first time.
