“枳实-白芍”药对治疗肝癌的网络药理学研究及分子靶点探讨

Study on the network pharmacology and molecular target of "Zhishi-Baishao" in the treatment of liver cancer

摘要: 探讨枳实-白芍发挥治疗肝癌的作用机制。利用网络药理学,从中药系统药理学分析平台筛选枳实-白芍活性成分及相关靶点蛋白;通过基因疾病关联数据库Genecard、OMIM数据库检索肝癌靶点,利用网络药理学分析平台及相关数据库对关键作用的节点进行生物功能和代谢通路分析,构建中药-疾病靶标蛋白相互作用网络图;最后在The Human Protein Atlas中寻找关键蛋白在癌组织与正常组织中的表达情况以及进行生存分析。结果筛选得出,枳实-白芍共有35个活性成分和65个共有靶点,GO分析结果涉及核受体功能、细胞外间隙、凋亡过程的负调控、染色质结合、RNA聚合酶Ⅱ启动子转录正调控和负调控等过程,KEGG结果显示与乙型肝炎、PI3-AKT、TNF等信号通路相关,PPI网络筛选出65个核心基因,关联度排名前三位分别是IL6、VEGFA、EGFR,三者的高低表达与肝癌患者五年生存率都有相关性。综上所述,枳实-白芍中的活性成分芍药苷、新橙皮苷、川橙皮素等通过癌症途径、PI3-AKT、TNF等信号通路对IL6、VEGFA、EGFR等相关靶点发挥转录正调控、核功能受体、凋亡途径等作用,为进一步实验研究提供了理论基础。

Abstract:

To explore the mechanism of Fructus Aurantii Immaturus and Radix Paeoniae Alba in the treatment of liver cancer.Using network pharmacology,the active components and related target proteins of Fructus Aurantii Immaturus and Radix Paeoniae Alba were screened from the pharmacology analysis platform of traditional Chinese medicine system;the target of liver cancer was retrieved from GeneCards and OMIM databases,and the biological functions and metabolic pathways of key nodes were analyzed by using the network pharmacology analysis platform and related databases to build the interaction between ztraditional Chinese medicine and disease target proteins Finally,the expression of key proteins in the human protein atlas was found in cancer and normal tissues,and survival analysis was carried out.The results showed that there were 35 active components and 65 common targets in Fructus Aurantii Immaturus and Radix Paeoniae Alba.The results of go analysis involved nuclear receptor function,extracellular space,negative regulation of apoptosis process,chromatin binding,positive regulation and negative regulation of RNA polymerase Ⅱ promoter transcription.KEGG results showed that it was related to hepatitis B,PI3-AKT,TNF and other signal pathways.Sixty-five core genes were screened by PPI network,IL6,VEGFA and EGFR were the top three in the correlation degree,and their high and low expression was correlated with the five-year survival rate of patients with liver cancer.In conclusion,paeoniflorin,neohesperidin and chuanhesperidin,the active components in Fructus Aurantii Immaturus,play the role of positive transcription regulation,nuclear function receptor and apoptosis pathway on IL6,VEGFA,EGFR and other related targets through cancer pathway,PI3-AKT,TNF and other signal pathways,providing a theoretical basis for further empirical research.
