摘要:

建立六月雪的UPLC指纹图谱和多成分含量测定方法,对不同产地六月雪药材的质量进行较全面的评价。通过对照品化学指认,并结合相似度评价、主成分分析、和正交偏最小二乘判别分析对16批六月雪样品的总体质量进行分析评价。16批六月雪样品指纹图谱具有14个共有峰,通过与对照品比对以及UPLC-ESI-QE-Orbitrap-MS定性分析指认了其中6个成分,分别为去乙酰车叶草苷酸、京尼平苷酸、车叶草苷酸、维采宁Ⅱ、槲皮素-3-O-α-L-鼠李糖基(1→2)-β-D-吡喃葡萄糖苷-7-O-α-L-鼠李糖苷和新绿原酸,各批样品与对照图谱的相似度均>0.900;多元统计分析结果表明16批六月雪样品可分成4类,确定了8个差异性标志物。16批次六月雪药材中去乙酰车叶草苷酸、车叶草苷酸和维采宁II的含量范围分别为2.035~4.422、1.210~3.279和0.087~0.288 mg/g,其中河南产区的六月雪样品中去乙酰车叶草苷酸和车叶草苷酸含量最高,而维采宁II含量偏低。该研究建立的方法简便、准确,可为不同产地六月雪药材的质量控制及其评价研究提供参考。

Abstract:

In this study,the ultra performance liquid chromatography(UPLC) was used to establish fingerprint and multi-component determination method of Herba Serissae,and provide a comprehensive evaluation of the drug from different habitats.The further quality assessment of the drug were carried out by chemical identification of reference substance combined with similarity evaluation,principal component analysis (PCA),and orthogonal partial least squares discriminant analysis (OPLS-DA).The UPLC fingerprint of Herba Serissae was established and fourteen common peaks were designated. Six common peaks were identified,namely deacetyl asperulosidic acid,neochlorogenic acid,geniposide,asperulosidic acid,vicenin Ⅱ and quercetin-3-O-α-L-rhamnopyranosyl(1-2)-β-D-glucopyranoside-7-O-α-L-rhamnopyranoside by comparing with the reference substances and UPLC-ESI-QE-Orbitrap-MS technical analysis.Multivariate statistical analysis showed that 16 batches of Herba Serissae were divided into four categories,and eight quality differential markers were determined.The content of deacetyl asperulosidic acid,asperulosidic acid and vicenin Ⅱ in 16 batches of Herba Serissae standard were decoction ranged from 2.035 to 4.422 mg/g,1.210 to 3.279 mg/g and 0.087 to 0.288 mg/g,respectively.The contents of deacetyl asperulosidic acid and asperulosidic acid ether in Herba Serissae of Henan province were the highest,while the content of vicenin Ⅱ was low.The method established in this study is simple and accurate,which can provide an quality control and evaluation for the Herba Serissae from different producing areas and provide reference.
