基于广泛靶向代谢组学分析花榈木不同部位挥发性代谢物差异

Differential analysis of volatile metabolites in different parts of Ormosia henryi by using widely-targeted volatilomics method

摘要:

花榈木(Ormosia henryi)是集药用、材用、景观于一体的重要珍贵植物。为进一步探究花榈木挥发性代谢物成分及不同部位的差异情况,采用气相色谱-质谱(GC-MS)广泛靶向代谢组学技术鉴定并分析了根、枝、叶代谢组分差异。结果表明,在根、枝和叶中共鉴定到589个挥发性代谢物,分为萜类、酯、杂环化合物、烃类等16类化合物。差异代谢物在倍半萜和三萜生物合成通路中显著富集。聚类热图结果表明,大多数代谢物在枝和叶中的富集量相对较高。萜类化合物在根中累积量降低,在枝和叶中富集,在叶中含量较高,其中柠檬烯、β-紫罗兰酮、α-蒎烯、α-松油醇等萜类成分累积量增加且具有重要生物活性化合物,具有较高利用价值。本研究对不同部位挥发性代谢物的分析为进一步探究花榈木不同部位药用特征成分和开发利用提供数据支撑。

Abstract:

Ormosia henryi is an important and precious plant which is applied to aspects of medicine,wood and landscape garden.In order to further explore the volatile metabolites composition and the differences among different parts of O. henryi,a broad targeted metabolomics technique was used to identify and analyze the differences of metabolic components and highly enriched components in roots,branches and leaves.The results showed that 589 volatile metabolites were identified and annotated in roots,branches and leaves,which were divided into 16 classes of compounds,such as terpenoids,esters,heterocyclic compounds and hydrocarbons.Differential metabolites are significantly enriched in sesquiterpene and triterpene biosynthetic pathways.The results of cluster heat map showed that the concentration of most metabolites in branches and leaves was relatively high.Terpene compounds were down-regulated in roots,enriched in branches and leaves,and had high content in leaves.Terpene compounds such as limonene,β-ionone,α-pinene and α-terpinol were up-regulated and had important bioactive compounds with high utilization value.The analysis of volatile metabolites from different parts of O.henryi provided data support for further exploration of the medicinal characteristics of different parts and development and utilization.
