基于UPLC-Q-TOF-MS/MS技术的江西特产中药茶芎化学成分研究

摘要:

茶芎是江西省九江地区的特产中药材之一,其药效物质基础尚不明确,故采用超高效液相色谱-四级杆-飞行时间串联质谱法(UPLC-Q-TOF-MS/MS)对茶芎的化学成分进行快速分析及鉴定。采用Titank C18(100 mm×2.1 mm,1.8 μm)色谱柱,以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,流速为0.25 mL/min,进样量为3 μL;选择电喷雾离子源分别在正、负离子模式下扫描分析,通过保留时间、精确相对分子质量和二级质谱裂解碎片,对所得成分的主要色谱峰进行鉴定。结果显示从茶芎中共鉴定出72个化学成分,主要包括苯酞类成分33个,有机酸类成分32个,生物碱类成分5个,其他类成分2个。该研究建立的UPLC-Q-TOF-MS/MS方法可以快速准确且较全面地分析鉴定茶芎中的化学成分,可为其质量控制及药效物质基础研究提供科学依据。

Abstract:

To rapidly analyze and identify the chemical composition of Chaxiong,one of the special Chinese medicine in Jiujiang area of Jiangxi Province,and its pharmacological substance basis is not clear,ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS /MS) was analyzed in this study.The chromatography was performed on a Titank C18 column (100 mm×2.1 mm,1.8 μm) with acetonitrile -0.1% formic acid water as the mobile phase for gradient elution with a flow rate of 0.25 mL/min and an injection volume of 3 μL.The mass spectrometry was scanned in positive and negative ion modes with electrospray ion source (ESI) to identify the main chromatographic peaks of the obtained components by retention time,accurate relative molecular mass and secondary mass spectrometry cleavage fragmentation.The results of this study showed that a total of 72 chemical components were identified from Chaxiong,including 33 phenanthrene components,32 organic acid components,five alkaloid components,and two other components.The UPLC-Q-TOF-MS/MS method established in this study can quickly,accurately and comprehensively analyze and identify the chemical components in Chaxiong,providing a reference for its quality control and pharmacological substance basis research.
