川木通的化学成分及鉴别研究

Chemical Constituents and Identification of the Caules of Clematis armandii Franch. and Clematis montana Buch.-Ham.

摘要:

研究川木通的化学成分,根据结果对中国药典川木通的薄层色谱鉴别内容提出商榷意见。采用硅胶柱、Sephadex LH-20分离及重结晶等方法,从川木通Clematis armandii Franch.的乙醇提取物中,分离纯化得到四种化合物。通过波谱数据、理化性质和对照参考文献,分别鉴定为9,12-octadecadienoic acid(Z,Z)-,2,3-dihydroxypropyl ester(1)、麦角甾醇(2)、豆甾醇(3) 和β-谷甾醇(4),其中1和2为首次分离得到。用TLC和HPLC图谱比对发现采集的11批川木通药材中均含有豆甾醇,而未发现齐墩果酸。中国药典以齐墩果酸为对照品薄层色谱法定性鉴别川木通的内容有待商榷。

Abstract:

Four compounds, 9,12-octadecadienoic acid(Z,Z)-,2,3-dihydroxypropyl ester (1), ergosterol (2), stigmasterol (3), and β-sitosterol (4), were isolated from the ethanol extracts of Clematis armandii Franch., and identified on the basis of the spectroscopic and physicochemical data with those reported previously. Compounds 1 and 2 were isolated from the stems of C. armandii for the first time. However, oleanolic acid, an indexed component in Chinese Pharmacopoeia for the identification of Clematis spp. was not detected in our conditions. To determine whether oleanolic acid presented in C. amandii and C. Montana, we collected 11 batch samples include nine for C. amandii and two for C. Montana. There was no trace of oleanolic acid in all 11 samples based on HPLC and TLC analysis, indicating the need for reinvestigation the chemical constituents and the indexed components of C. spp..
