南海深海细菌Bacillus amyloliquefaciens GAS 00152抗菌代谢产物研究

摘要: 采用多种色谱分离技术,从南海深海细菌Bacillus amyloliquefaciens GAS 00152发酵液中分离得到12个化合物,经波谱数据分析分别鉴定为4-苯基-3-丁烯酰胺(1)、苯乙酰胺(2)、苯乙酸-2-(4-羟苯基)乙酯(3)、对羟基苯乙醇(4)、环(甘氨酸-2-氨基丁酸)(5)、环(甘氨酸-亮氨酸)(6)、环(甘氨酸-L-脯氨酸)(7)、环(D-脯氨酸-L-缬氨酸)(8)、环(4-羟基脯氨酸-亮氨酸)(9)、环(N-甲基甘氨酸-苯丙氨酸)(10)、环(L-2-羟基脯氨酸-苯丙氨酸)(11)、环(2-哌啶酸-苯丙氨酸)(12)。其中,化合物1为新天然产物。测试化合物1~12 对番木瓜炭疽菌和香蕉黑星菌的抑制活性,发现化合物9对两种热带水果致病菌显示出中等抗菌活性。

Abstract: A bacterium strain, GAS 00152, was isolated form marine sediment collected at a depth of 2476 m in the South China Sea. This bacterium was identified as Bacillus amyloliquefaciens by analysis of its 16S rRNA gene sequence. Chemical investigation of the fermentation broth of GAS 00152 was carried out using silica gel column chromatography, PTLC and semi-preparative HPLC. Twelve compounds were isolated and identified as 4-phenyl-3-butenamide (1), 2-phenylacetamide (2), phenylacetic acid-2-(4-hydroxyl phenyl) ethyl ester (3), 4-hydroxyphenethylethanol (4), cyclo-(Gly-Leu) (5), cyclo-(Gly-Leu) (6), cyclo-(Gly-L-Pro) (7), cycl-(D-Pro-L-Val) (8), cyclo-(4-hydroxy-Pro-Leu) (9), cyclo-(N-methyl-Gly-Phe) (10 ), cyclo-(L-2-hydroxy-Pro-Phe) (11) and cyclo- (pipecolinyl-phenylalanine) (12) on the basis of spectroscopic data and comparison with literatures. Among them,Compound 1 was a new natural product. Compounds 1-12 were evaluated for their antibacterial activity. Compound 9 showed moderate activities against C. papayae and P. musrarum.
