响应曲面优化枇杷叶绿原酸超声微波提取工艺

Optimization of Ultrasonic-Microwave Extraction of Chlorogenic Acid from Eriobotrya japonica Leaves by Response Surface Methodology

摘要: 利用响应曲面法对枇杷叶中绿原酸的提取工艺进行优化。以单因素试验为基础,根据Box-Behnken中心组合设计原理,选取乙醇体积分数、料液比、微波功率和微波时间4因素和3水平进行响应曲面分析,建立绿原酸提取率的二次多项数学模型,分析各因素的显著性和交互作用,得到绿原酸提取工艺的最佳条件为:乙醇体积分数70%、料液比1:15、微波功率400 W、微波时间5 min,绿原酸提取率为5.84%。

Abstract: In this study,response surface analysis was applied to optimize the extraction process of chlorogenic acid from Eriobotrya japonica. On the basis of single-factor experiments,the Box-Behnken center composite experimental design principle was used to design the experiments and the response surface analysis with 4 factors and 3 levels was adopted.A second order quadratic equation for extraction of chlorogenic acid was created.After analyzed the significance of various factors and their interactions,the optimal extraction conditions for the extraction ofchlorogenic acid from E.japonica leaves were determined as follows:70% aqueous ethanol as the extraction solvent,1:15 as sample-to-solvent ratio,50 ℃ as extraction temperature,ultrasonicationfor 30 minand 5 minmicrowavewith 400w as microwave power.The extraction yield of chlorogenic acidwas determined to be 5.84% under the optimized extraction conditions.
