摘要: 采用循环伏安法和紫外光谱法研究了羧甲基壳聚糖-Cr(III)配合物与鲱鱼精DNA之间的作用方式。结果发现:(1)羧甲基壳聚糖-Cr(III)的存在导致Fe(CN)63-/4-探针分子氧化还原峰电流下降,式量电位正移,显示羧甲基壳聚糖-Cr(III)和Fe(CN)63-/4-与DNA之间存在竞争性作用,说明羧甲基壳聚糖-Cr(III)通过插入方式与DNA相互作用的;(2)中性红的加入导致羧甲基壳聚糖-Cr(III)/DNA体系的特征吸收峰发生增色效应、峰位红移并伴有等吸收点出现,这进一步表明羧甲基壳聚糖-Cr(III)是以插入方式与鲱鱼精DNA发生了相互作用,导致DNA分子的构象变化,并且CMC-Cr与DNA的结合比n(CMCCr):n(DNA)为3:1。

Abstract: The interaction between carboxymethylchitosan-Cr (III) Complex (CMC-Cr) and herring sperm DNA were investigated by cyclic voltammetry and UV-Vis spectra.The results indicated:Firstly,the peak current of probe molecule Fe(CN)63-/4- at Au/DNA electrodes obviously decreased and the peak potential shifted positively because of the existences of CMC-Cr,which showed that there was a competitive effect between CMC-Cr and Fe(CN)63-/4- with herring sperm DNA,and revealed that a binding mode intercalation was interacted between CMC-Cr and herring sperm DNA.Secondly,the absorption intensity of CMC-Cr/DNA system increased with the adding of neutral red (NR),the maximum absorption peak had a red shifted and appeared equal absorption point,which further illustrated that a binding mode intercalation was interacted between CMC-Cr and herring sperm DNA,thus led to the change of DNA conformation.The combining ratio of n(CMC-Cr) and n(DNA) was 3:1.
