高效液相色谱法考察木绣球不同部位中绿原酸含量

Determination of Chlorogenic Acid in Different Parts of Viburnum macrocephalum by HPLC

摘要: 为比较木绣球不同部位中绿原酸的含量,采用索氏提取法提取木绣球花、花托、茎、叶等四个部位的绿原酸并用HPLC法测定含量。色谱条件如下:Eclipse XDB-C18色谱柱(4.6 mm × 250 mm,5 μm),流动相为乙腈-0.1%磷酸水溶液(10:90),检测波长为327 nm,流速为1.0 mL/min。在上述色谱条件下,绿原酸在0.34~6.80 μg/mL范围内线性良好。结果表明木绣球不同部位中绿原酸含量为:花托(1.14%)>花(1.02%)>茎(0.51%)>叶(0.45%)。木绣球中花托部位绿原酸含量最高,木绣球整体均具有潜在的研究与开发价值。

Abstract: The contents of chlorogenic acid in flowers,torus,stems and leaves of Viburnum macrocephalum was determined by HPLC. V.macrocephalum was extracted by Soxhlet extraction method.The chromatographic condition was established as follows:Eclipse XDB-C18 (4.6 mm × 250 mm,5 μm) column,the mobile phase composed of acetonitrile-0.1% phosphoric acid aqueous solution (10:90),the detection wavelength was 327 nm,the flow rate was 1.0 mL/min.Under the developed HPLC conditions,good linear relationship was established within the range of 0.346.80 μg/mL.The content of chlorogenic acid in different parts of V.macrocephalum was in following order:torus (1.14%)>flowers (1.02%)>stems (0.51%)>leaves (0.45%).The torus of V.macrocephalum had the highest content of chlorogenic acid. V macrocephalum had the potential exploitation value.
