蚯蚓谷胱甘肽还原酶分离提取的研究

Extraction and Purification of Glutathione Reductase from Earthworm Eisenia fetida

摘要: 本研究利用闪式提取与超声提取相结合的方法从蚯蚓中提取纯化了谷胱甘肽还原酶(GR),利用正交试验对提取条件进行了优化,并对GR的酶学性质进行了研究。结果表明,GR的最佳提取条件为:pH值7.5,料液比1:5,闪提时间45 s,超声时间15 min。提取的GR比活力达到655.9 U/mg pr.,纯化了32.3倍,回收率为58.4%。利用SDS-PAGE电泳测得蚯蚓GR的分子量为110 kDa。GR的最适pH值和温度分别为8.0和50 ℃。

Abstract: Glutathione reductase (GR) is an enzyme that plays important physiological role and widely distributes throughout organisms.GR catalyzes reaction to turn the oxidized glutathione (GSSG) into the reduced glutathione (GSH).In this study,GR was purified to electrophoretically homogeneous state from earthworm Eisenia fetida. The purification process involved flash extraction,ultrasonic extraction,acetone precipitation,ammonium sulfate precipitation and ion exchange chromatography.The results showed that the optimal extraction conditions determined by orthogonal experiments were:pH of 7.5,solid/liquid ratio of 1:5,extraction time of 45 s,and ultrasonic treatment time of 15 min.Under these conditions,GR was purified 32.3-fold to a specific activity of 655.9 U/mg pr.,and was obtained with a yield of 58.4%.The molecular weight of the purified enzyme was estimated 110 kDa by SDS-PAGE.The optimal pH and temperature were 8.0 and 50 °C for GR.
