PI3K/Akt/GSK-3β信号通路在6-姜酚保护PC12细胞对抗β淀粉样蛋白诱导细胞凋亡的作用

Protective Effect of 6-Gingerol on PC12 Cells Induced by Amyloid -β-Protein by PI3K/Akt/GSK-3β Signaling Pathway

摘要: 研究6-姜酚对β淀粉样蛋白(A-β1-42)引起的PC12细胞凋亡的保护作用及其可能的分子信号通路。通过MTT检测不同浓度A-β1-42对PC12细胞的作用及不同浓度6-姜酚对A-β1-42诱导细胞凋亡的保护作用,采用蛋白免疫印迹法检测6-姜酚对A-β1-42诱导的PC12细胞糖原合成激酶3β(GSK-3β)、磷酸化GSK-3β(Ser9),Akt及磷酸化Akt(Ser473)表达的影响。结果显示6-姜酚能显著减少A-β1-42引起的细胞凋亡;PC12细胞经A-β1-42作用48 h后,80及120 μM 6-姜酚预处理组中GSK-3β及Akt的磷酸化水平则均显著高于A-β1-42处理组的。说明6-姜酚对抗A-β1-42引起的细胞毒性作用,可能与其激活Akt的活性、抑制GSK-3β的活性有关。

Abstract: To investigate the protective effects and the molecular signaling pathways of 6-gingerol against damage of PC12 cells induced by Amyloid β-protein (Aβ1-42). MTT assay was also performed to determine the protective effects of 6-gingerol (40,80,120,200,and 300 μM) on PC12 cells apoptosis induced by Aβ1-42. Western blot was performed to evaluate the expressions of glycogen synthase kinase-3β (GSK-3β),phosphorylated GSK-3β (Ser9),Akt and p-Akt (Ser473). Results showed that 6-gingerol significantly attenuated apoptosis induced by Aβ1-42 in PC12 cells.In addition,6-gingerol (80 and120 μM) pretreatment obviously enhanced the expressions of p-GSK-3β and p-Akt compared with the Aβ1-42 treatment group.6-Gingerol might protect PC12 cells against apoptosis induced by Aβ1-42  via suppressing the activation of GSK-3β and enhancing the activation of Akt.
