UPLC-MS测定不同产地及不同部位昆明山海棠中6种活性成分含量

摘要: 建立了超高效液相色谱-质谱联用同时测定昆明山海棠中雷公藤吉碱、雷公藤次碱、雷公藤甲素、去甲泽拉木醛、雷酚内酯、雷公藤内酯甲6种活性成分含量的方法,并对不同产地及不同部位药材含量进行了分析。采用Phenomenex Luna C18色谱柱(50×2.0 mm,3 μm),以乙腈和0.1%甲酸水为流动相梯度洗脱,流速0.3 mL/min,柱温30 ℃,质谱条件为电喷雾电离源(ESI),多反应检测模式(MRM)。雷公藤吉碱、雷公藤次碱、雷公藤甲素、去甲泽拉木醛、雷酚内酯、雷公藤内酯甲分别在6.80~1360.00、5.90~1180.00、1.20~240.00、25.05~5010.00、1.45~290.00、1.65~330.00μg/L浓度范围内显示出良好的线性关系(r≥0.9998),平均加样回收率为98.19%~100.91%,RSD为1.46%~2.99%。应用该方法对来自不同产地及不同部位的36个昆明山海棠样品进行了测定,该方法简便、快速、准确,可用于昆明山海棠药材的质量控制。

Abstract: An ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) method was developed for the simultaneous determination of wilforgine,wilforine,triptolide,demethylzeylasteral,triptophenolide and wilforlide A in different habitats and different parts of Tripterygium hypoglaucum Hutch.The chromatographic separation was performed on a Phenomenex Luna C18 column (50×2.0 mm,3 μm) with gradient elution consisting of acetonitrile and 0.1% formic acid in water at a flow rate of 0.3 mL/min.The column temperature was set at 30 ℃.Detection was carried out on a triple quadrupole tandem mass spectrometry by multiple-reaction monitoring (MRM) and an electrospray ionization (ESI) was operated in positive mode.All calibration curves of the 6 active compounds showed good linearity in the range of 6.80-1360.00,5.90-1180.00,1.20-240.00,25.05-5010.00,1.45-290.00 and 1.65-330.00μg/L (r≥0.9998).The recoveries were in the range of 98.19%-100.91% and RSDs were between 1.46% and 2.99%.The method was applied for the determination of 6 compounds in 36 samples of T.hypoglaucum Hutch from different habitats and different parts.The results indicated that this method was simple,rapid and reliable.It can be used for the quality control of T.hypoglaucum Hutch.
